Sperm osteopontin mRNA expression levels and its correlation on semen quality and fertility in Madura bulls. Biodiversitas 24: 563-570. Osteopontin (OPN) gene transcripts influence spermatogenesis and germ cell development. Therefore, transcriptome analysis is needed to identify the fertility factor OPN in Madura bull sperm. Madura cattle are crosses originating from Bos indicus (zebu) and Bos javanicus (banteng). This study aims to examine sperm osteopontin (OPN) mRNA expression levels and its correlation with semen quality and fertility in Madura bulls. Frozen semen samples from Madura bulls were categorized as high-fertile (n: 4, average field conception rate: 78.28±3.25%) or sub-fertile (n: 4, average field conception rate: 66.73±5.01%). In post-thaw semen samples, sperm motility, viability, membrane, acrosome, and DNA fragmentation index were evaluated. OPN expression in sperm total RNA was analyzed using Real-Time Quantitative Polymerase Chain Reaction (RT-qPCR). The data analysis between the two fertility groups was assessed with Student's t-test and Pearson Square. Receiver-operating characteristic analysis and diagnostic sensitivity and specificity calculations were performed. Sperm motility, acrosome intact, and DFI differ (p<0.05) between groups, whereas viability and membrane plasma intact have no significance (p>0.05). In high-fertile Madura bulls, OPN mRNA was upregulated (p<0.05). The OPN mRNA expression had a strong correlation with the field conception rate (R: 0.807, P<0.05), sperm motility, and intact acrosome (R: 0.899, p<0.01; R: 0.804, p<0.05) respectively. In contrast, OPN negatively correlated with sperm DFI (R: -0.764, p<0.05). The OPN predicted bull fertility with 81.3% accuracy, 75% sensitivity, and 75% specificity. Thus, OPN may serve as a potential biomarker of Madura bull fertility.

Semen volume is the amount of ejaculate released by male Bali cattle during one ejaculation, which consists of spermatozoa and seminal plasma. The purpose of this study was to determine the effect of Bali cattle ejaculation volume on the quality of fresh and frozen semen. Semen collection was carried out using an artificial vaginal collection method. The collected semen was then evaluated with microscopic parameters: motility, viability, and abnormalities. Semen came from 7 male Bali bulls. The data obtained were analyzed by one-way ANOVA. The results showed that there was no significant difference (P>0.05) between the volume of Bali cattle ejaculation on the quality of fresh semen and also post thawing based on the parameters tested. The sperm concentration of Bali bulls has average 935.75 ± 80.25 x 106. The average percentage of fresh semen motility was 70.79±1.26 while that for frozen was 47.83±1.89. The average viability of fresh spermatozoa was 80.04±3.68 while frozen 52.87±0.67. The average abnormality value of fresh spermatozoa was 8.54±1.88 while frozen was 23.83±4.22. Based on these results, it was concluded that the different volume of ejaculate of Bali cattle did not affect the quality of fresh semen of Bali cattle, both fresh and frozen based on the parameters of motility, viability, and abnormalities.

Effects of Adding Glutathione to AndroMed Diluent on Intact Plasma and Acrosome Membranes, and Progressive Motility of Cattle Spermatozoa During Freezing Processes

Adding endogenous antioxidants to the diluent is significantly associated with semen quality during the freezing process. This study aimed to investigate the effects of adding glutathione to AndroMed diluent on the preservation of crucial sperm attributes, namely, intact plasma membrane (IPM), intact acrosome membrane (IAM), and progressive motility of Bali cattle spermatozoa. A completely randomized design was used, and spermatozoa samples were obtained from a Bali cattle and divided into two diluent treatment groups (36 diluent samples in each group with six replications), namely pure AndroMed as the control and a group with the addition of glutathione (1 mmol) to AndroMed. Each treatment was replicated six times and evaluated at three freezing stages, including post-dilution, post-equilibration, and post-thawing, for crucial sperm properties. The results indicated that fresh Bali cattle spermatozoa had progressive motility, IAM, and IPM of 75%, 89%, and 88%, respectively. During the freezing process, there was a significant decrease in semen quality, including progressive motility, IAM, and IPM of spermatozoa after dilution to post-equilibration and post-equilibration to post-thawing in both treatment groups. Meanwhile, the addition of 1 mmol of glutathione to AndroMed diluent had a significant difference in increasing progressive motility, IAM, and IPM of Bali cattle spermatozoa at each stage of semen freezing, including post-dilution, post-equilibration, and post thawing when compared with controls. Based on the results, it can be concluded that adding 1 mmol of glutathione to the AndroMed diluent enhanced the quality and integrity of Bali cattle semen, including progressive motility, IAM, and IPM during the freezing process.
