Beneficial Effects of Montelukast against Cisplatin-Induced Acute Renal Damage in Rats

Objective: In this study, the therapeutic and protective effects of montelukast against cisplatin (CP)-induced acute renal damage were investigated. Materials and Methods: Thirty-five female rats were divided into five groups as follows: (1) control, (2) montelukast (10 mg/kg daily for 10 days per-oral (p.o.), (3) CP (single dose 7 mg/kg intraperitoneally (i.p.)), (4) CP + montelukast (10 mg/kg daily for 10 days p.o., after 3 days of the injection of CP), (5) montelukast (10 mg/kg daily for 10 days p.o.) + CP (single dose 7 mg/kg i.p., after the last dose of montelukast). At the end of the experiment, malondialdehyde (MDA), a lipid peroxidation product, myeloperoxidase (MPO), and reduced glutathione (GSH) levels were determined in the renal tissue. Also, blood urea nitrogen (BUN) and creatinine (Cr) levels were assayed from the trunk blood samples. Results: CP treatment caused a significant elevation of MDA, MPO, BUN, and Cr levels when compared with the control group. Also, GSH levels were found to be reduced due to the CP treatment. Montelukast administration after CP injection ameliorated all of these parameters. Our histopathological findings (marked swelling of epithelial cells, tubular dilatation, tubular desquamation, and loss of brush border in the kidney) were consistent with the biochemical results. Conclusion: Montelukast treatment after CP injection exerted therapeutic effects against CP-induced acute kidney damage.

Thirty male rats were randomized into five groups. The animals received ticagrelor 5 mg/kg, 10 mg/kg and 20 mg/kg or normal saline 0.1 mL/kg (control) orally before the procedure. The shame group only had laparatomy. An ischemia-reperfusion injury was done by clamping the renal hilus. There was less malondealdehyde assay (MDA) in ticagrelor groups than the control group, and this decrease was statistically significant (P=0.001 in all ticagrelor received groups). The glutathione peroxidase (GPx) and glutathione reductase (GR) were elevated in ticagrelor groups at all doses. Similar findings were observed in all treatment groups. The 5 mg ticagrelor treated group had no changes in glomerules and tubules relative to the control group via histology. Dilated tubular structures were similar to the sham group-both at 10 and 20 mg ticagrelor. Caspas-3 and NF-KB activity was similar in ticagrelor treated groups to sham group. Our study showed that ticagrelor is an effective agent to protect kidney from renal ischemia-reperfusion injury. Ticagrelor may protect the tissues by reducing the concentration of reactive oxygen species and inducing the antioxidant system-especially with the pleiotropic effect. Bu amaçla, 30 erkek rat rastgele olarak 5 gruba ayırılmıştır. Bu hayvanlara, yapılacak işlemlerden önce ticagrelor 5 mg/kg, 10 mg/kg ve 20 mg/kg dozlarında, kontrol grubuna da 0.1 mL/kg serum fizyolojik oral olarak verilmiştir. Sham grubunda ise sadece laparatomi yapılmıştır. İskemi-reperfüzyon hasarı renal hilusun klemplenmesi yoluyla yapılmıştır. Ticagrelor verilen gruplarda saptanan malondialdehit (MDA) düzeyinin kontrol grubundan daha az olduğu ve bu düşüklüğün istatistiksel olarak anlamlı olduğu tespit edildi (tüm ticagrelor grupları için P=0.001). Abstract. Testicular torsion or torsion of the spermatic cord is one of the most serious urological conditions. It causes testicular injury, which potentially leads to male subfertility. The turning of the spermatic cord and spermatic structures around themselves results in biochemical and histological changes; however, following testicular detorsion, tissues undergo reperfusion that causes more severe damage than that induced by ischemia. Since the primary causes of testicular damage are reactive oxygen species production, an increase in intra-mitochondrial calcium concentration and an increased rate of cellular apoptosis, different medications may potentially be effective. It seems that several medications, experimentally and sometimes clinically, serve an adjuvant role in the cellular damage that occurs following ischemia-reperfusion. Antioxidants, calcium channel blockers, phytotherapeutical medicinals, anaesthetics, hormones and platelet inhibitors may potentially create a solid basis for an adjuvant restoring therapy and ameliorate testicular function following torsion. The current study aimed to review the relevant literature and discuss the actions of a number of molecules that may protect the testes during ischemia/reperfusion injury.
