Molecular and phylogenetic analysis of bovine coronavirus based on the spike glycoprotein gene

Abstract: Bovine coronavirus has been associated with diarrhoea in newborn calves, winter dysentery in adult cattle and respiratory tract infections in calves and feedlot cattle. In Cuba, the presence of BCoV was first reported in 2006. Since then, sporadic outbreaks have continued to occur. This study was aimed at deepening the knowledge of the evolution, molecular markers of virulence and epidemiology of BCoV in Cuba. A total of 30 samples collected between 2009 and 2011 were used for PCR amplification and direct sequ… Show more

“…In addition, all the Argentinean strains sequenced grouped separately from Mebus reference strain, which has been used for systematic vaccination of Argentinean cattle (Dr. Parreño, personal communication). Furthermore, Argentinean strains form a well-defined cluster, separate of the other geographical clusters like most of the Brazilian strains as was previously observed (Bidokhti et al, 2012;Chung et al, 2011;Fulton et al, 2013;Martinez et al, 2012;Takiuchi et al, 2008). The length of the branch indicates Argentinean cluster divergence from all the other groups of BCoVs.…”

“…In contrast, little epidemiological information is available regarding BCoV detection, incidence and characterization in cattle from Central and South American countries. In Cuba, BCoV sequences clustered with BCoV strains from USA, suggesting a common origin for these viruses (Martinez et al, 2012). In South America, most of the information comes from studies conducted in Brazil (Asano et al, 2010;Barros et al, 2013;Brandao et al, 2008Brandao et al, , 2006Takiuchi et al, 2008).…”

“…To determine the genomic sequence of nsp12, S, and, N genes, a set of 17 overlapping RT-PCR products, from 553 to 1260 nucleotides, were generated using a total of 42 primers (Lau et al, 2011;Hasoksuz et al, 2002;Martínez et al, 2012). RT-PCR reactions were performed using One Step RT-PCR System (Qiagen, Holden, Germany), from 2.5 μl of nucleic acid in a final volume of 25 μl, according the manufacturer's instructions.…”

“…An additional maximum likelihood tree based on 143 complete sequences of S gene of BCoV, BCoV-like, and HCoV-OC43 from USA, Brazil, South Korea, China, Italy, Swedish, Denmark and France was constructed. To this end, 81S gene sequences from BCoVs and BCoVs-like were added to the previous S gene alignment (Martínez et al, 2012;Beaudeau et al, 2010;Decaro et al, 2009a;Jeong et al, 2005;Zhang et al, 1991;Park et al, 2007;Chung et al, 2011;Park et al, 2006). The accession numbers corresponding to these sequences are provided in Tables S2 and S3 (supplementary material).…”

“…Thus, the unique evolution of BCoV in yaks may be related to the special geographical environment of the Qinghai-Tibet Plateaui.e. the high-altitude (average altitude of over 4000 m), low oxygen, low temperature and low atmospheric pressurewhere the yak live, as well as to the characteristics of the host species [28][29][30][31].…”
