MMP2 and MMP9 are Associated with Apical Periodontitis Progression and Might be Modulated by TLR2 and MyD88

Abstract: The aim of this study was to evaluate the expression of MMP2 and MMP9 during apical periodontitis (AP) progression in TLR2 (TLR2 KO) and in MyD88 (MyD88 KO) knockout mice compared to wild type (WT) mice. AP was induced in mandibular first molars of TLR2 KO (n= 18), MyD88 KO (n= 18), and WT mice (n= 18). After 7, 21, and 42 days, the animals were euthanized and the jaws were dissected and subjected to histotechnical processing. Subsequent sections were stained by immunohistochemistry and evaluated for detection… Show more

“…The increase levels of glucose in the blood and changes in inflammatory molecules cause alterations in the periodontal tissue [36], thereby modulating the activation of different kinases and proteases involved in the development of periodontitis [16]. Recent studies have demonstrated an association between MMP-2 and periodontitis in patients without diabetes [9, 11, 12, 19, 39]. Nevertheless, the relationship between periodontitis and the gelatinolytic activity of MMP-2 in T2DM patients is contradictory [23, 24].…”

“…During periodontal tissue inflammation, overexpression of MMPs leads to an increase in MMP levels in biological fluids such as saliva and gingival crevicular fluid [18, 19]. Previous studies on this topic have found higher levels of MMP-2 in saliva from patients with periodontitis [20, 21].…”

“…It is highlighted that the present findings must be exercised with caution and not transferred directly to clinical conditions because of obvious limitations of immunohistochemical analysis and studies in animals. Regarding the semi-quantitative analysis, the score system was used as in several previous published studies (15,30,31). Also, most of evaluated molecules (ALP and OPN) are extracellular proteins, making it difficult to perform a counting of immunolabelled cells.…”

“…Control slides in which the primary antibody was omitted were used to test the specificity of immunostaining. Positive staining in periapical tissues was scored (1= mild, 2= moderate, and 3= intense) as previously described (23). For this analysis, all slides were prepared in the same batch to obtain a standardized staining and were evaluated by an experienced blind examiner.…”
