Microtiter Tests for Detecting Antibody in Bovine Serum to Parainfluenza 3 Virus, Infectious Bovine Rhinotracheitis Virus, and Bovine Virus Diarrhea Virus

Abstract: Microneutralization tests for detection of antibody in bovine serum to three bovine viruses are described. The Madin-Darby bovine kidney cell line was used with parainfluenza 3 virus (PI 3), whereas serially cultivated bovine embryonic kidney cells were used for infectious bovine rhinotracheitis virus and bovine virus diarrhea virus. Comparison of micro-hemagglutination-inhibition (HI) with micro-serumneutralization (SN) tests for PI 3 showed the SN test to be more sensitive, more specific, and therefore more … Show more

“…All serum samples were tested using serum neutralization test for detection of specific neutralizing antibodies against all vaccinal viruses of Pneumo-4 commercial and prepared oil adjuvants. SNT applied in micro-titration plate technique following the procedure described by Rossi and Kiessel, (1971). The Serum Neutralizing Antibody titers of the tested serum samples were expressed as log10 of the reciprocal serum dilution that protects >50% of micro titration plate dilution wells, and these following the calculation formula of Reed and Muench, (1938).…”

“…Antibodies to B. abortus were identified by plate agglutination (Alton et al, 1988). Sera were heat-inactivated at 56°C for 60 min and 2-fold dilutions were tested for end-point neutralizing antibodies to National Animal Disease Laboratory (NADL; USA) reference isolate of BVDV and LA (Los Angeles) reference strain of BHV-1 using a standard microtitration procedure (Rossi and Kiesel, 1971).…”

“…The serum neutralization tests for BHV-1 and BVDV were performed following the already described methods (ROSSI and KIESEL, 1971). The viral strains were: LA of BHV-1, and NADL of BVDV.…”
