MicroRNA miR-330-3p suppresses the progression of ovarian cancer by targeting RIPK4

Abstract: Previous studies reported that miR-330-3p was involved in the progression of several cancers, but the potential roles of miR-330-3p in ovarian cancer (OC) were unclear. In the current study, we aimed to explore the expression pattern and functions of miR-330-3p in OC. The expression level of miR-330-3p in OC tissues and cell lines was detected using RT-qPCR. The proliferation, migration and invasion of OC cells were detected using CCK-8 assay and transwell assay, respectively. Bioinformatics analysis and lucif… Show more

“…The DNA sequence, which was amplified, was cloned into pmirGLO dual-luciferase vector (Promega, Madison, WI, USA) for building reporter vectors of wild type (WT) OIP5-AS1, mutant type (MUT) OIP5-AS1, wild type (WT) SIRT1-3ʹUTR and mutant (MUT) SIRT1-3ʹUTR, respectively. We positioned NR8383 cells in 24-well plates overnight and applied Lipofectamine 2000 (Invitrogen, USA) to transfect the above luciferase reporter vectors and miR-128-3p mimics or miR-NC into the cells [ 29 ]. Forty-eight hours after transfection, the luciferase activity test was conducted using a dual-luciferase reporter gene assay system (Promega, Madison, WI, USA).…”

“…Then, the membranes were incubated with anti-VEGF (ab150375; Abcam), anti-endoglin (ab252345; Abcam), and anti-GAPDH (ab8245; Abcam) overnight at 4 °C. Following the incubation of secondary antibody for 1 h. The protein signals were analyzed by the ECL system kit (Pierce, Rockford, USA) [ 26 ].…”

“…The CCK-8 assay was carried out as previous observation [ 23 ]. Briefly, at 24 h after transfection, Cal-27 cells were grown in 96-well plates at 37°C and cultured for 24 h. Then, 10 μL of CCK-8 solution was added to each well, and the plates were incubated another 2 h. Absorbance at 450 nm was measured.…”
