Mesenchymal stem cells derived exosomes and microparticles protect cartilage and bone from degradation in osteoarthritis

Abstract: Mesenchymal stem or stromal cells (MSCs) exert chondroprotective effects in preclinical models of osteoarthritis (OA). Most of their therapeutic effects are mediated via soluble mediators, which can be conveyed within extracellular vesicles (EVs). The objective of the study was to compare the respective role of exosomes (Exos) or microvesicles/microparticles (MPs) in OA. MPs and Exos were isolated from bone marrow murine BM-MSCs through differential centrifugation. Effect of MPs or Exos was evaluated on OA-lik… Show more

“…The therapeutic effect of MSC‐derived exosomes on arthritis have also been investigated. In several studies, intravenous or intra‐articular exosomes isolated from MSCs were shown to inhibit macrophage activation, inflammation, and cartilage destruction in in vitro and in vivo models via inhibition of catabolisis, inflammatory markers and apoptosis, increase of collagen type II synthesis, and modulation of T and B cells …”

“…Extracellular vesicle (EV), including exosomes, may also be a novel therapeutic option for OA. Studies in small animal OA models showed that stem cell derived EVs attenuated OA disease progression in vitro and in vivo by displaying similar biological functions as stem cells; however, further investigations into the mechanism is required before application to the clinic . These are exciting times for stem cell biology, both in the lab and the clinic, to help in the understanding and management of OA.…”

“…Clinical and animal studies demonstrated the efficacy of MSC therapies and the importance of MSC paracrine secretion in various diseases [27,28]. Cosenza et al injected MSC-derived exosomes into a collagenase-induced OA model and found that it protected mice from joint damage [29]. Zhang et al investigated if intra-articular injection of human embryonic MSC-derived exosomes repaired and regenerated osteochondral defects in a rat model [30].…”
