The conversion of an amine to an olefin by elimination of the nitrogen atom and an adjoining hydrogen atom is a useful procedure for degradation and synthesis. The Hofmann exhaustive methylation method has been used most often to bring about change, but other methods as the thermal decomposition of amine oxides and the pyrolysis of amine phosphates or acetyl or benzoyl derivative have often been employed to advantage. This chapter reviews the Hofmann elimination because of its extensive history. Alternative methods are considered.

Von S-haltigen Steroiden sind solche mit Schwefel in 3-Stellung am haufigsten beschriebenl) ; nur in zwei Patenten2) werden Steroide mit S in 17-Stellung dargestellt. Die Einfiihrung von zwei S-Atomen ist fur die Positionen 1.5, 2.3 und 1.7 bekannt3). Wir beschreiben im folgenden die Darstellung von 3.17-Dimercapto-steroiden der 5a-Androstan-Reihe.Ausgehend von 3a-oder 3P-Hydroxy-Scc-androstanon-(l7) (1,6) erhielten wir nach bekannten Verfahren iiber die 3-Tosylate 2 und 5 die 3-Thiocyanato-Derivate 4 bzw. 3. Reduktion der daraus mit HCI/HzS entstandenen 17-Thioketone 7,8 mit Lithiumalanat ergab die gewiinschten 3.17-Dithiolq die sich als kristallisierte Dibenzoate 9, 10 gewinnen lieBen. Die sterische Anordnung der Endprodukte an C-3 ist durch die SN~-Substitution derTosylate gegeben: 3a-OH+3a-OTos+3P-SCN-+3~-SAcyl bzw. 3P-OH+~-+3cc-SAcyl.In Analogie zur Reduktion von 17-Carbonylgruppen4) und da die Reduktion von Thiocarbonylgruppen entsprechend verlauft la), schreiben wir den 17-SH-Gruppen P-Konfiguration zu.In Verfolgung eines zweiten Weges stellten wir aus Scc-Androstandion-(3.17) mit HCl/ H2S das 3-Thion-17-on 11 dar, das sich in das 17-Diathylmercaptdl 12 iiberfiihren lieB. Dessen LiAlH4-Reduktion lieferte jedoch lediglich harziges Material. der Konstitutionsermittlung.
