Background and Objective: The current study aimed to evaluate cardiovascular risk factors, haematological and biochemical parameters, and serum endocan concentrations in lichen planus (LP) patients. Methods: This study was conducted with 86 cases, including 43 LP patients and 43 healthy controls. Cardiovascular risk factors, haematological and biochemical parameters, and endocan levels were evaluated. Results: The serum endocan concentrations of LP patients were not significantly different from those of the healthy controls (p > 0.05). There were no significant differences in the serum endocan levels according to classification by cardiovascular risk factors and smoking history (p > 0.05). In the LP group, white blood cell count, platelet distribution width and monocyte count/high-density lipoprotein cholesterol ratios were significantly higher when compared to the healthy controls (p < 0.05). The LP group had a lower mean platelet volume than the healthy controls (p < 0.05). Conclusions: Serum endocan levels did not change significantly in patients with LP, and there were significant differences in haematological and biochemical parameters.

ÖZAmaç: Pigmente purpurik dermatoz (PPD) peteşi ve hiperpigmente maküllerle karakterize bir hastalıktır. Hastalığın etiyoloji tam olarak bilinmemektedir. Ortalama trombosit hacmi (OTH) trombosit fonksiyon ve aktivasyonunu gösteren bir belirteçtir. Trombositler hemostaz ve koagulasyondaki rollerine ek olarak inflamasyon ve immün sistem üzerinde de önemli rol oynarlar. Biz bu çalışmada, PPD ve OHT arasındaki olası ilişkiyi araştırdık. Yöntemler: PPD tanısı olan 40 hasta çalışmaya dahil edildi. Kontrol grubu sağlıklı 64 kişiden oluşturuldu. Çalışma dizaynı retrospektifdi. Yaş, cinsiyet, OTH ve trombosit düzeyleri hasta dosyalarından kayıt edildi. Trombosit fonksiyonunu etkileyen herhangi bir kronik hastalığı ve ilaç kullanımı olan olgular ça-lışma dışı bırakıldı. Bulgular: Ortalama OTH düzeyleri hasta grubunda 9,7±2,1fl ve kontrol grubunda 8,0±1,1 fl idi. Bu farklılık istatistiksel olarak anlamlıydı (p<0,001). Ortalama trombosit düzeyi hasta grubunda 246,2 ± 55,8 ve kontrol grubunda 261,6± 49,6 idi. Her iki grup arasında trombosit düzeylerinde anlamlı bir farklılık yoktu (p=0,169). Sonuç: Biz bu çalışmamızda OTH ile PPD arasında anlamlı bir ilişki bulduk. Hastalığın patogenezinde artmış trombosit aktivasyonun olası rolünü belirlemek için daha kapsamlı ve ileri çalış-malara ihtiyaç vardır. Anahtar Kelimeler: Pigmente purpurik dermatoz, trombosit, ortalama trombosit hacmi ABSTRACT Objective: Pigmented purpuric dermatosis (PPD) is a chronic skin disease characterized by petechial and pigmentary macules. The etiology of the disease remains unknown. Mean platelet volume (MPV) is a marker of platelet (PLT) function and activation. PLTs play important roles in inflammation and innate and adaptive immunity in addition to their roles in hemostasis and thrombosis. In this study, we investigated the possible association between MPV and PPD. Methods: Forty patients with PPD were enrolled in the study. The control group consisted of 64 healthy subjects. The study design was retrospective. The age, gender, and MPV and PLT levels were recorded from patients' charts. Cases who had received any medicine affecting PLT function and had a concomitant disease were excluded from the study. Results: The mean MPV levels were 9.7±2.1 fl and 8.0±1.1 fl in the patient and control groups, respectively. This difference was significant (p<0.001). The mean PLT levels were 246.2±55.8 and 261.6±49.6 in the patient and control groups, respectively. There was no significant difference in the PLT counts between the two groups (p=0.169). Conclusion: In this study, we found a significant correlation between MPV and pigmented purpuric dermatosis. There is a need for more detailed and prospective studies to determine the possible role of increased PLT activation in the pathogenesis of the disease.

Objective: This study aimed to explore the effects of STAT3 targeting by let-7a on T-cell proliferation and IFN-γ secretion in psoriasis. Methods: From January 2013 to January 2015, 40 patients with psoriasis (psoriasis group) and 38 volunteers undergoing plastic surgery (control group) were enrolled in this study. Pearson correlation analysis was performed to evaluate the correlation between let-7a and STAT3 expression. T-cells were isolated and subjected to different transfection methods. A dual luciferase reporter assay was carried out to confirm STAT3 as a target gene of let-7a. Let-7a, STAT3 and IFN-γ mRNA expression was detected by quantitative real-time fluorescent polymerase chain reaction (qRT-PCR), and pSTAT3 protein levels were determined by Western blot. T-cell proliferation was evaluated with a cell counting kit-8 (CCK-8) assay. Results: The level of STAT3 mRNA and pSTAT3 was higher, but let-7a expression was lower in the psoriasis group than the control group. Pearson correlation analysis indicated that STAT3 expression was negatively correlated with let-7a expression. T-cells transfected with inhibitors exhibited greater IFN-γ mRNA expression and T-cell proliferation than transfected T-cells and T-cells transfected with a non-sense sequence, while T-cells transfected with let-7a mimics exhibited lower IFN-γ mRNA expression and T-cell proliferation than transfected T-cells and T-cells transfected with a non-sense sequence. This suggested that siRNA-STAT3 could reverse the increase in IFN-y mRNA expression and T-cell proliferation induced by let-7a inhibitors. Conclusion: Our results demonstrated that let-7a inhibits T-cell proliferation and IFN-γ secretion by down-regulating STAT3 in psoriasis.
