Matrine‐Type Alkaloids from the Seeds of Sophora alopecuroides and Their Potential Anti‐inflammatory Activities

Abstract: Three new matrine‐type alkaloids, 8β‐hydroxyoxysophoridine (1), 9β‐hydroxysophoridine (2), 9β‐hydroxyisosophocarpine (3), together with one known analog, 11,12‐dehydromatrine (4), were isolated from the seeds of Sophora alopecuroides L. The structures of new compounds were elucidated using extensive spectroscopic techniques including the experimental and calculated ECD data. The anti‐inflammatory activities of all the isolates on NO production in RAW 264.7 cells stimulated by lipopolysaccharide were evaluated.… Show more

“…The results of immunosuppression experiments with cyclosporine as a positive control showed that compound 18 has a strong inhibitory activity. In the structure-activity analysis of the compounds, it is assumed that dichlorocyclopropane fused to the D ring and trichloromethyl bonded to C-13 had a negative effect on the activity, which is probably due to high steric hindrance Compounds 20-22 are hydroxymatrine, reported in 2021, whose hydroxyl group is attached to the 8 or 9 positions of the B ring [46]. The experimental results of the inhibitory activity of the three compounds on the production of NO induced by LPS in RAW 264.7 cells show that compound 20 has a significant inhibitory effect.…”

“…[ 1 ] The seeds of S. alopecuroides (known as “Ku‐Dou‐Zi” in China) were used as traditional Chinese medicine for the treatment of many kinds of illnesses such as fever, rheumatism, bacterial infection and inflammatory diseases. [ 2‐3 ] Previous phytochemical and pharmacological investigations of the S. alopecuroides extracts revealed that matrine‐type alkaloids are one of principal active chemical constituents, [ 4‐7 ] which are responsible for diverse intriguing bioactivities such as antiviral, [ 8 ] anti‐tumor, [ 9‐10 ] and promising anti‐inflammatory activities. [ 3,10 ] It's noteworthy that matrine is used as an adjuvant drug in the treatment of COVID‐19 with good reputation, [ 11‐12 ] which was supported by network pharmacology, molecular docking analyses, [ 13 ] as well as clinical trials.…”

“…S. alopecuroides is an important medicinal plant widely used in pharmaceutical preparations, such as Kexieling tablets, fuyan suppositories, S. alopecuroides oil liniment, and so on [ 16 ]. The whole plant can be used as medicine, with antitumor [ 17 , 18 , 19 ], anti-inflammatory [ 20 , 21 ], antiviral [ 22 ], insecticidal [ 15 , 23 ], and other pharmacological effects. Studies have shown that S. alopecuroides has a well-developed root system and strong drought and salinity resistance [ 15 , 24 ] and can fix nitrogen [ 25 ].…”
