Mapping of the SLA complex class III region by pulsed field gel electrophoresis

Abstract: The fine order of genes in the class III region of the swine major histocompatibility complex (MHC), the SLA complex, was examined by pulsed field gel electrophoresis (PFGE) and Southern blot analysis. Four genes, C2, HSP70, TNF alpha, and CYP21, were analyzed. The CYP21, C2, and HSP70 genes were all located within a 200-kb NotI fragment. The C2, HSP70, and TNF alpha genes cohybridized to a 420-kb SalI fragment. The TNF alpha gene is linked to the class I region by a 390-kb NotI fragment. Combined with a previ… Show more

“…We should also mention that in the mouse H-2 d complex the Bat-1 gene is duplicated in association with the D a and L d class I genes (Wroblewski et al 1994), whereas in pig and human only one BAT1 gene seems to exist. The comparison of our PGFE data with those published by others in miniature pigs (Xu et al 1992;Wu et al 1995) are generally in agreement, with some differences with a few restriction enzymes, which may indicate restriction enzyme site polymorphisms between miniature and Large White breeds. The distance between the class II region (DRA) and the class III region, which is about 200 kb in length, seems to be conserved between humans, pigs, and mice (Lunney 1994).…”

“…The swine MHC region also comprises three closely linked Hsp70 loci which have been mapped to the class III region [34,35,50]. Linkage of the Hsp70 gene with MHC has also been reported in the goat and cattle [5,13].…”
