Currently, very few male contraceptive methods are available. Areca nuts are known to have an antifertility effect, but the effective, safe dose is not certainly known. Data on its effectiveness and safety are needed to develop the potential of the areca nuts as an herbal contraceptive in men. This study determines the antifertility effect of areca nut administration at doses of 40, 30, and 20 mg/kgBW and their safety on the kidneys and liver. The green-colored raw areca nuts were used. The treatment groups were male rats whose fertility status was known before treatment. The control group were given distilled water. The treatment were given one dose a daily dissolved for 28 days of treatment, then male rats were bred again. Rats were terminated for histopathological examination of liver, kidneys, testes, and for analysis of spermatozoa. The administration of areca nuts caused a decreasing number of litters, but only the group given 40 mg/kgBW dose had a significant decrease (p<0.05). There were a damaged testes, a decreasing number and motility of spermatozoa as well as increasing spermatozoa abnormalities. There were no changes in liver tissue, but there was a mild grade cast in kidney tissue. Areca nut have antifertility effect in male rats with dose dependent manner, histopathological examination revealed cast nephotoxic but not hepatotoxic.

With persistently high global rates of unintended pregnancy and contraceptive nonuse, nonadherence and discontinuation, new contraceptive methods must address the needs of women and men who seek alternatives to their current options. Methods under development aim to reduce potential side effects, improve access and ease of use, ensure safety, increase secondary benefits associated with method use and expand options for both women and men. Developmental approaches employed to enhance current methods utilize new delivery systems and novel active pharmaceutical ingredients. This will improve overall user satisfaction with the methods used while expanding the number of options available to provide choice and value user autonomy in the highly diverse contraceptive markets around the world.

Background Lactate dehydrogenase C (LDHC) is specifically expressed in male germ cells and plays critical roles in glycolysis. Glycolysis is required to supply energy for sperm motility. Previous studies showed that Ldhc knock‐out mice exhibit impaired sperm motility. Objectives We established human LDHC knock‐in (hLDHC KI) mice and examined whether hLDHC KI mice can be used to assess LDHC‐targeting drugs. Material and methods HLDHC was knocked‐in to the mouse Ldhc (mLdhc) allele using the CRISPR/Cas9 system. Mating tests, sperm motility examinations with a computer‐assisted sperm analysis (CASA) system, and in vitro fertilization (IVF) were performed. Furthermore, the effect of an LDH inhibitor was analyzed with CASA and IVF. Results HLDHC was detected at the protein level in hLDHC KI spermatozoa. hLDHC KI mice exhibited comparable sperm motility and male fertility to wild‐type (WT) mice. When we performed IVF using the LDH inhibitor more specific to hLDHC than mLDHC, fertilization rates were reduced in hLDHC KI mice but not in WT mice. Discussion and conclusion Our results reveal that hLDHC can rescue the absence of mLDHC. Differences in the effect of the LDH inhibitor between WT and hLDHC KI mice indicate that hLDHC KI mice can be a good model to assess hLDHC inhibitors for preclinical contraceptive studies.
