Background. The study of the effect of end products of protein and nucleic acid metabolism on the immune system is of interest not only theoretical but also practical in line with the problems of urotoxins and hemodialysis. In previous studies, we found significant links between uricemia and uricosuria, on the one hand, and immune parameters, on the other hand. In this study, the spectrum of nitrogenous metabolites was expanded due to creatinine, urea and bilirubin. Material and methods. Experiment was performed on 60 healthy female Wistar rats. The plasma level and urinary excretion of the nitrogenous metabolites were determined. Immune status was assessed by thymocytogram, splenocytogram, blood leukocytogram and immunocytogram, as well as phagocytosis of blood neutrophils and monocytes. Results. Both negative and positive metabolic-immune correlations were revealed. Calculation of multiple correlation coefficients between individual metabolite parameters and constellations of immune parameters revealed the maximum immunotropic effect of uricosuria (R=0,637). This is followed by excretion of urea (R=0,617) and creatinine (R=0,606), bilirubinemia (R=0,589), creatinineemia (R=0,567), uricemia (R=0,566) and plasma urea level (R=0,500). The canonical correlation between the constellation of nitrogenous metabolites, on the one hand, and the parameters of immunity, on the other hand, was very strong: R=0,921; χ 2 (154)=282; p<10 -6 . Conclusion. Nitrogen metabolites exhibit significant immunotropic activity, both suppressor and enhancing.

Previously, we found a wide range of uric acid metabolism parameters and functional relationships of uricemia and uricosuria with the parameters of immunity in healthy female rats analyzed. It was found by canonical correlation analysis that both parameters of uric acid metabolism determine the immunity status of rats by 71%. The purpose of this study is to determine the features of immune status in rats with quantitatively and qualitatively different uric acid metabolism. Material and Methods. Experiment was performed on 60 healthy female Wistar rats 220-300 g. Among them 10 animals remained intact, using tap water from drinking ad libitum. The rats of others groups for 6 days administered through the tube various fluids. The serum and urine levels of the uric acid (uricase method) were determined. In the blood, the parameters of immunity were determined. From thymus and spleen made smears-imprints for counting splenocytogram and thymocytogram. For them, as well as immunocytogram and leukocytogram of blood, Shannon's entropy was calculated. Results. Screening of immune profiles of four quantitative and qualitative uric acid metabolism clusters revealed 6 functionalmetabolic patterns, two of which are quasilinear (enhancing and suppressing), with immune extremes at moderately elevated uricemia, two U-shaped with immune optimum at moderately reduced uricemia levels, as well as a non-reactive pattern. Discriminant analysis revealed 14 parameters of immunity, the constellation of which four clusters of metabolism of uric acid clearly distinguish. Conclusion. Endogenous uric acid exerts a modulatory immunotropic effect in healthy female rats.168

Why Hyperuricemia Does Not Necessarily Induce Gout?

Hyperuricemia is a risk factor for gout. It has been well observed that a large proportion of hyperuricemia individuals have never had a gout attack(s), while some patients with gout can have a normuricemia. This raises a puzzle of the real role of serum uric acid (SUA) in the occurrence of gout attacks. As the molecule of uric acid has its dual effects in vivo with antioxidant property as well as being an inflammatory promoter, it has been placed in a delicate position in balancing metabolisms. Gout seems to be a multifactorial metabolic disease and its pathogenesis should not rely solely on hyperuricemia or MSU crystal. This critical review aims to unfold the mechanisms of the SUA role participating in gout development. It also discusses some key elements which are prerequisite for the formation of gout in association with the current therapeutic regime. The compilation should be helpful in precisely fighting for a cure of gout clinically and pharmaceutically.
