Macrophage-Derived IL-1β Regulates Emergency Myelopoiesis via the NF-κB and C/ebpβ in Zebrafish

Abstract: Myeloid phagocytes, neutrophils in particular, are easily consumed when they fight against a large number of invading microbes. Hence, they require efficient and constant replenishment from their progenitors via the well-orchestrated emergency myelopoiesis in the hematopoietic organs. The cellular and molecular details of the danger-sensing and warning processes to activate the emergency myelopoiesis are still under debate. In this study, we set up a systemic infection model in zebrafish (Danio rerio) larvae v… Show more

“…Macrophage-derived il1β actives nf-κb and c/ebpβ, two fast-reactive transcription factors, rapidly. In an LPS-induced systemic infection zebrafish model, the macrophage is the first cell type to detect LPS and secret il1β, which is further responded to by MPCs, rather than HSPCs, expanding myelopoiesis through the synergistic cooperation of nf-κb and c/ebpβ (Wei et al 2020 ).…”

“…Similarly, inhibition of the components of the IL1β pathway, such as caspase-1, reduces leukocyte recruitment to the wound and also NFκB activity [198]. IL1β also regulates infection-driven emergency myelopoiesis via NF-κB and C/ebpβ [203]. Finally, both TNFα and IL1β play important functions in regeneration [52,85,204].…”

“…Cytokines exert a complicated influence on hematopoietic output; however, the actual effects of cytokines tend to be dose-dependent and time-dependent. The dose and acute or chronic exposure may bring opposite influences ( 53 , 54 , 109 ), which has been observed in IL-1 ( 45 , 110 112 ), TNFα ( 109 ), IFNs, and TGF-β ( Table 2 ). Besides the direct influence on HSCs, specific cytokines can also interfere with the normal signals controlling hematopoietic output downstream of stem cells.…”
