FASN Gene Polymorphism in Indigenous Cattle Breeds of Turkey

Abstract: The aim of this study was to determine the variants of the thioesterase (TE) beta-ketoacyl reductase (KR) domains of the Fatty Acid Synthase (FASN) gene, in the East Anatolian Red (EAR) and South Anatolian Red (SAR) cattle breeds. It has been suggested that the FASN gene is effective on fatty acid composition of meat in cattle. In this study, the genotype and allele frequencies of g.17924 A>G, g.18440 G>A and g.16024 G>A, g.16039 T>C in TE and KR domains, respectively, were detected by using polymerase chain r… Show more

“…FASN rs41919985 in the population studied was polymorphic with two genotypes, namely, AG and GG. Different from the results of this study, in which the AA genotype was not found, a previous study found three genotypes (GG, AG, and AA), but the AA genotype only appears at a low frequency of 0.06 in Turkey Anatolian Red cattle [26] and 0.08 in Chinese Holstein cattle and Fleckvieh bulls [18,27]. The AA genotype was reported at a higher frequency (0.28) in Dutch Holstein cattle [9].…”

“…This process is catalyzed by FASN [27]. SNP rs41919985 is found on exon 39 of the FASN gene [26] and is a missense mutation that results in an amino acid change from threonine (ACC) to alanine (GCC). This mutation likely affects the structure of the substrate-binding site and therefore, may result in differences in enzymatic regulation and different milk compositions [26,27].…”

“…As reported in other studies, this SNP has an effect over the fatty acid composition of adipose tissue (Zhang et al, 2008;Li et al, 2012;Oh et al, 2012;Yeon et al, 2013;Oztabak et al, 2014), and on fat deposition, as found in Korean cattle by Oh et al (2018), who commented that this SNP could be a casual mutation for increasing fat deposition.…”

“…The FASN g.17924A>G (T2264A), SNP is located in the thioesterase domain of FASN enzyme and made a change from polar, neutral, and hydrophilic to nonpolar, aliphatic, and hydrophobic in this motif, affecting the structure of the substrate binding site and therefore changing the specific activity of thioesterase domain towards C14-acyl ACP (Acyl carrier protein domain), which transfers the growing fatty acid chain throughout the different domains of this enzyme (Oh et al, 2018;Zhang et al, 2008), affecting its affinity with the fatty acid (Oztabak et al, 2014). Therefore, the thioesterase domain determines the carbon chain length of the final product of this enzyme and any change or variation in this domain could affect the fatty acid composition of meat and fat (Zhang et al, 2008).…”

“…Amplification was performed in the thermocycler (MasterCycler -Eppendorf, USA). Oztabak et al (2014) About 1μl of each PCR product was diluted in 10μl of denaturing buffer (98% formamide, 20mM EDTA, 0.05% bromophenol blue and 0.05% xylene cyanol), denatured at 95°C for 5min and maintained on ice. The total volume was applied onto 12.5% polyacrylamide gel.…”
