Ligustrazine Phosphate Ethosomes for Treatment of Alzheimer’s Disease, In Vitro and in Animal Model Studies

Abstract: Abstract. In the present study, we have investigated transdermal administration of ligustrazine phosphate (LP), as an antioxidant, for the treatment of Alzheimer's disease (AD). The LP transdermal ethosomal system was designed and characterized. Franz-type diffusion cells and confocal laser scanning microscopy were used for the in vitro permeation studies. Furthermore, the effect of LP transdermal ethosomal system on AD was evaluated in the scopolamine-induced amnesia rats by evaluating the behavioral performa… Show more

“…The application was first put forward by Touitou in 1996(Touitou, 1996 and was specially tailored to efficiently deliver various molecules into deep skin layers and across the skin (Ainbinder et al, 2010). Ethosomes has been studied for delivery of various drugs ranging from low molecular weight drugs such as acyclovir , minoxidil (Jun-Bo et al, 2007), tacrolimus (Li et al, 2012), ligustrazine phosphate (Shi et al, 2012), econazole nitrate (Verma & Pathak, 2012), ibuprofen (Shumilov et al, 2010) to polypeptides and gene medicines (Godin & Touitou, 2003).…”

“…With regard to cognition enhancing function, TMP has been shown to improve learning and memory ability in scopolamine-or D-galactose-lesioned rodents (Zhang et al, 2004;Shi et al, 2012). Although these insults provide animal models of memory deficit, there are few literatures referring to the protective potential of TMP in more physiologically relevant models of Alzheimer's disease.…”

“…Also, ethosomes can be used to increase the bioavailability of natural products [24] . For instance, in previous studies, ethosomes were used to enhance penetration of ligustrazine [25] and alkaloids from Sophora alopecuroides [26] . Thus, this drug delivery system is likely to increase the penetration of QC through the skin.…”
