Lectin histochemical study of lipopigments present in the cerebellum of Solanum fastigiatum var. fastigiatum intoxicated cattle.

Abstract: S. fastigiatum var. fastigiatum was dried, ground and administered to 2 cattle by ruminal cannula at 5 g/kg daily for 5 days a week during 107 and 140 days, respectively. At the end of the study, the animals were sacrificed and cerebellum samples were obtained for histological study. It was shown that there was Purkinje cell vacuolization in the treated animals. The Purkinje cells showed high binding affinity for lectins from Concanavalia ensiformis (Con-A) and succinyl-WGA (sWGA). Staining was also evident fo… Show more

“…The decrease in locomotor and rearing frequencies caused by S. guaraniticum extract administration suggests depressant and/or motor disturbance. This effect is similar to symptoms of cattle intoxication caused by Solanum species (Rech et al, 2006;Riet-Correa et al, 1983;Paulovich et al, 2002), which includes locomotive incoordination and loss of equilibrium. Although this study does not present microscopic evaluation of brain tissues, these results should be further investigated prior to indication of this plant as a phytotherapeutic agent.…”

“…Yet there is paucity of pharmacological and toxicological data on such S. guaraniticum properties. Previous studies of a similar species, S. fastigiatum in high doses, have evidenced liver toxicity (Sabir, Rocha, 2008), which was also related to bovine intoxication, affecting the central nervous systems of the animals (Rech et al, 2006;Riet-Correa et al, 1983;Paulovich et al, 2002).…”

“…ConA binds to a-mannosyl residues and is specific for a-D-mannose and a-D-glucose, while the positive staining with WGA and S-WGA suggest the presence of terminal moieties consisting of b-D-N-acetyl-glucosamine and acetyl-neuraminic acid (Damjanov, 1987). Stored material with S. fastigiatum intoxication reacted strongly with ConA, sWGA, WGA and RCA-I and based on this lectin-binding pattern it was proposed that this intoxicosis represents a glycolipid storage disease (Paulovich et al, 2002). The pattern of lectin staining in maldronksiekte partially agrees with those observed in intoxication with Sida carpinifolia (Driemeier et al, 2000), locoweed (Astragalus lentiginosus) and swainsonine (Swainsona galegifolia) (Alroy et al, 1985).…”

“…Estudos de microscopia eletrônica revelaram que os vacúolos dos neurônios de Purkinje na intoxicação por S. fastigiatum correspondem a inclusões membranosas lipídicas semelhantes às encontradas nas neurolipidoses de animais e humanos (Barros et al 1987). Isso parece ter sido confirmado por estudos histoquímicos com lectinas, que sugerem que a intoxicação é uma doença de depósito glicolipídico (Paulovich et al 2002).…”
