Lead-induced oxidative stress and metabolic alterations in Cassia angustifolia Vahl.

Abstract: Forty-five-days old plants of Indian senna (Cassia angustifolia Vahl.) were subjected to 0 -500 µM lead acetate (Pb-Ac) in pot culture. Changes in contents of thiobarbituric acid reactive substances (TBARS), ascorbate, glutathione, proline, sennosides (a+b), and activities of superoxide dismutase (SOD), ascorbate peroxidase (APX), glutathione reductase (GR), and catalase (CAT) were studied at pre-flowering (60 d after sawing, DAS), flowering (90 DAS) and post-flowering (120 DAS) stages of plant development. “…Pb is known to induce oxidative damage to higher plants (Qureshi et al 2007). In this study, the enhancement of H 2 O 2 and MDA concentrations in A. macrocephala showed that the oxidative stress and the peroxidation of membrane lipids were induced by Pb treatment (Figs.…”

“…Its increasing levels in soil environment inhibit seed germination and root growth of maize seedlings (Eun et al 2000). In addition, it is known to induce oxidative stress that cause damage to the biomolecules such as chloroplasts, soluble protein, proline, ascorbate, enzymes, etc (Verma and Dubey 2003;Qureshi et al 2007;Zhao et al 2009;Shu et al 2012).…”

“…This is mostly universal oxide reductase that scavenges H 2 O 2 via a two-electron transfer producing O 2 and H 2 O (Lin and Kao, 2000). The major function of CAT is to metabolize the peroxide liberated in the peroxisome following the conversion of glycollate during photorespiration (Qureshi et al, 2007). In the present study, in shoots of Raphanus sativus, the decrease in CAT activity under Pb concentration indicating removal of H 2 O 2 and toxic peroxides and, in turn, the reduction in the free radical mediated lipid peroxidation under Pb toxicity.…”

“…However, with increasing Pb level, reduction in CAT activity was observed, which has been attributed to the activation of the enzyme protein (Feieraband and Engel, 1986) and the decrease in enzyme synthesis (Mittler, 2002). The decrease in CAT activity could indicate its inactivation by accumulation of H 2 O 2 induced by lead in Cassia angustifolia (Qureshi et al, 2007).…”

“…The increased SOD levels lead to the over-production of H 2 O 2 . H 2 O 2 production during oxidative stress in plants is removed by CAT enzymes (Qureshi et al 2007). The probable greater activity of CAT in A. cepa suggests its larger role in the removal of H 2 O 2 (Nocter et al 2002).…”
