Laser Enhanced Combinatorial Chemo-photothermal Therapy of Green Synthesis Gold Nanoparticles Loaded with 6Mercaptopurine on Breast Cancer Model

Abstract: Purpose We tend to evaluated a method for loading 6Mercaptopurine (6MP) on green synthesized hybrid chitosan gold nanoparticles (AuNPs) forming 6MP-AuNPs nanocomposite for the first time in combinatorial chemo-photothermal therapy. Methods The AuNPs were synthesized using chitosan as a reducing and capping agent. Different concentrations of 6MP were mixed AuNPs. Cells were incubated with 6MP and 6MP loaded AuNPs for 48 h and then exposed to laser. … Show more

“…Moreover, the better electrostatic attraction of DOX with AuNPs than citrate groups might be accountable for the extra band at longer wavelengths. It has been revealed theoretically and experimentally that aggregation of AuNPs leads to additional plasmon absorption at higher wavelengths when nanoparticles are electronically attached to each other [ 16 ]. Consequently, violet color of the DOX-AuNPs solution as well as the UV-Vis spectrum slightly unchanged after 72 h storage in DMEM at room temperature, indicating the formation of a stable particle suspension, as shown in Fig.…”

“…Generally, nanocarriers are non-specifically internalized into cells via endocytosis or phagocytosis compared to the passive diffusion mechanism of free drugs into the cell [ 65 ]. Positively charged nanocarriers like 6MP-CNPs and 6MP-CNPs-AuNPs were known to facilitate their penetration through the negatively charged cancer cell membrane [ 66 ]. So, the electrostatic interaction could enhance the absorption of our proposed nanocarriers by MCF7cells.…”

“…The color of the AuNPs' suspension was dark purple. The narrow band of the curve corresponds mostly to the monodisperse nature of AuNPs, without any aggregation and agglomeration [20,21]. TEM observations were employed to clarify the morphology of formed AuNPs; the particles are spherical with approximate size (12 ± 2 nm) with uniform size distribution.…”
