Lanthanum Chloride Inhibits LPS Mediated Expressions of Pro-Inflammatory Cytokines and Adhesion Molecules in HUVECs: Involvement of NF-κB-Jmjd3 Signaling

Abstract: Background/Aims: To investigate the regulation of LaCl3 on lipopolysaccharides (LPS)-induced pro-inflammatory cytokines and adhesion molecules in human umbilical vein endothelial cells (HUVECs). Methods: Primary cultured HUVECs were pretreated with 2.5 µM LaCl3 for 30 min followed by 1 µg/ml LPS for 2 h. Pro-inflammatory cytokine and adhesion molecule expressions were determined by real-time RT-PCR and ELISA. NF-κB/p65 nuclear translocation was examined by immunofluorescence and immuno-bl… Show more

“…Protein was extracted from tissues with commercial lysis buffer. Western blotting was performed as described previously 17. Primary antibodies against zonula occluden (ZO)-1 (Invitrogen, Carlsbad, California, USA), ZO-2 (Abcam, Cambridge, UK), occludin (Invitrogen), claudin-4 (Abcam) and glyceraldehyde 3-phosphate dehydrogenase (GAPDH, Invitrogen) were used.…”

“…Cellular inflammatory stress has been shown to stimulate excessive or aberrant cytokine and adhesion molecule expression, which induces endothelial dysfunction [4, 5]. Angiotensin II (AngII) activates the transcription factor nuclear factor kappa-B (NF-κB), which induces tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6), ICAM-1, VCAM-1, and E-selectin expression, prompting vascular injury [6].…”

“…NF-κB has been shown to regulate ICAM-1 expression in various cell types [33] , [34] . Indeed, we proved that P. aeruginosa -induced ICAM-1 protein and mRNA levels and monocyte adhesion were inhibited by the inhibitor of NF-κB (HLN) or siRNA of p65 or p50 ( Fig.…”
