Kinetic characterization and comparison of various protein crosslinking reagents for matrix modification

Abstract: We have characterized the relative efficacies of a number of protein crosslinking agents that have the potential for use in the crosslinking of proteinaceous matrices both in vitro and in vivo. The crosslinkers tested were; l-threose (LT), Genipin (GP), Methylglyoxal (MG), 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide hydrochloride (EDC), proanthrocyanidin (PA) and glutaraldehyde (GA). The relative effectiveness of the crosslinkers with regard to their saturating concentrations was: GA > PA > EDC > MG = GP >>… Show more

“…However, PC crosslinking is due to hydrogen bonds, not covalent bonds, resulting in crosslinking with hydroxyproline in ECM proteins and maintaining a soft and pliable tissue [9,[21][22][23] . Proline is an efficient hydrogen bond acceptor, so PC has an extremely high affinity for proline-rich proteins [9,22,[33][34][35][36] . ECM proteins collagen and elastin are proline-rich, which promote PC to form hydrogen bonds [22,23] .…”

“…12 In recent years, it has been used as an effective reagent to crosslink ECMs of bovine precardium and porcine heart valve by forming hydrogen bond between hydroxyl of PC and proline residual in collagen and elastin. 9,13 Inspired by the advantages of PC, we supposed that PC may be used for crosslinking the ECMs of decellularized vessels. Therefore, the aim of this study was to explore PC as a useful reagent for crosslinking saphenous vein ECM (svECM) substitutes for small-diameter blood vessels to overcome the shortcomings of non-or GA-crosslinked vascular ECM substitutes.…”
