Vanda tricolor is an orchid endemic to Mount Merapi whose existence is threatened by the eruption of Mount Merapi which can occur at any time and the community has taken it directly from nature. Several attempts have been made to preserve the Vanda tricolor orchid, one of which is through in vitro culture. In vitro culture studies of Vanda tricolor have been carried out and large numbers of shoots have been obtained through multiplication and then raised by subculture. Plantlets obtained must be acclimatized to adapt and be returned to their original habitat. Growing medium and fertilizer application are important factors in the acclimatization process. This study aims to determine the best type of medium and fertilizer for acclimatization of Vanda tricolor orchids. This research was conducted using an experimental method in the preparation of a Factorial Completely Randomized Block Design. The first factor is the type of medium, namely coconut husk, moss and coconut husk + moss, the second factor is the type of fertilizer, namely without fertilizer, foliar fertilizer and liquid organic fertilizer. From the two treatment factors, 9 treatment combinations were obtained. Each treatment was repeated 3 times and each replication consisted of 3 samples. The results showed that there was a mutual influence between the type of medium and fertilizer. The coconut husk + moss is the best medium for acclimatization of Vanda tricolor orchids. Meanwhile, foliar fertilizer and liquid organic fertilizer compared to without fertilization, can increase the growth of Vanda tricolor orchids during acclimatization.

Interspecific hybridization of Dendrobium mirbelianum x D. nindii or D. discolor, in vitro seed germination, seedling growth and plantlet acclimatization. Biodiversitas 24: 3004-3011. Dendrobium mirbelianum Gaudich. (P1), D. nindii W.Hill (P2), and D. discolor Lindl. (P3) as a parent crossing are potentially resulting a new type of orchid as the novel breeding achievement. This study aimed to identify orchid crossing compatibility and evaluate the most effective and efficient media for hybrid seed germination and growth. The study consisted of three experiments, namely interspecific hybridization (experiment 1), in vitro hybrid seed germination (experiment 2), and in vitro growth of the hybrid seedlings (experiment 3). The first experiment was conducted by a simple modified line×tester mating design, and all experiments used a completely randomized design, with five replications in the first experiment and three replications in the second and third experiments. The first experiment presented all of the crossing emerging seed pods, but only hybrid seeds from cross D. mirbelianum x D. nindii and its reciprocal cross that potent to germinate in all tested media as the second experiment result. The third experiment showed FF+tryptone was the best media indicated by the highest and heaviest seedling after 8 months after sowing. All of the crossings resulted seed pods, however, only D. mirbelianum x D. nindii and its reciprocal cross that able to germinate in all tested media. Foliar fertilizer is the most recommended media for germinating hybrid orchid seeds, and FF+tryptone is the best for seedling growth.

Anggrek Dendrobium merupakan salah satu tanaman hias yang bernilai ekonomis tinggi. Minat masyarakat pada anggrek khususnya jenis Dendrobium cukup tinggi dan mendominasi pasar sehingga kebutuhan bibit anggrek jenis ini juga semakin tinggi. Tahapan kritis dalam menentukan keberhasilan pertumbuhan bibit anggrek juga ditentukan oleh teknik aklimatisasi. Perbedaan kondisi lingkungan pada tahap aklimatisasi dapat menyebabkan planlet mengalami transpirasi berlebihan dan mengganggu penyerapan nutrisi. Kitosan sebagai bahan organik alami merupakan turunan sederhana dari kitin yang dapat berfungsi sebagai biofertilizer dan bioimmuner. Tujuan penelitian ini untuk mengetahui respon pertumbuhan bibit anggrek terhadap pemberian kitosan untuk keberhasilan tahap aklimastisasi. Metode penelitian menggunakan Rancangan Acak Lengkap (RAL) dengan satu faktor yaitu konsentrasi kitosan. Tahap aklimatisasi dengan menngunakan aplikasi kitosan dengan konsentrasi 0 ppm, 2 ppm, 2,5 ppm, 3 ppm, 3,5 ppm, 4 ppm, 4,5 ppm dan 5 ppm. Data yang diperoleh selanjutnya dianalisis ragam dengan uji F pada taraf 5%. Perlakuan yang berpengaruh nyata diuji lanjut dengan Duncan Multiple Range Test (DMRT) pada taraf kepercayaan 95%. Hasil penelitian menunjukkan bahwa pemberian kitosan dengan konsentrasi 3 ppm berpengaruh nyata dalam meningkatkan parameter presentase tumbuh tanaman, tinggi tanaman, jumlah daun, lebar daun, panjang daun, panjang akar, jumlah akar, dan berat segar tanaman anggrek Dendrobium sonia pada tahap aklimatisasi.
