Molecular cytogenetic analyses using fluorescence in situ hybridization (FISH) and genomic in situ hybridization (GISH) were carried out to elucidate inter-specific relationships among wild Lilium species distributed in Korea. FISH revealed four to eight 45S rRNA gene loci, which are located on chromosomes 1-7, 10, and 11 among the different species. In contrast, the 5S rRNA gene locus was conserved on the long arm of chromosome 3, occasionally with two adjacent sites on the same chromosome arm in a few species. The 5S rDNA site was located adjacent to the 45S rDNA site in only three species, Lilium distichum, Lilium hansonii, and Lilium tsingtauense. GISH analysis using genomic DNA probes detected strong hybridization of genomes between diploid and triploid Lilium lancifolium species, demonstrating that triploid plants were derived from diploid L. lancifolium and not from Lilium maximowiczii. Phylogenetic analysis of the ITS and NTS sequences supported the cytogenetic data as well as Comber's classification of the genus Lilium.

The detailed karyotypes of diploid (2n = 2x = 24) and triploid (2n = 3x = 36) of L. tigrinum were constructed based on fluorescence in situ hybridization (FISH) using 5S rDNA and 45S rDNA probes. The lengths of mitotic metaphase chromosomes ranged from 15.92 to 30.18 um, with a total length/genome of 250.46 um in diploid, and 15.24 to 28.16 um, with a total length/genome of 233.42 um in triploid. The chromosome composed of two pairs of metacentrics (Chromosomes 1 and 2), four pairs of subtelocentrics (Chromosomes 3, 4, 5, and 6) and six pairs of telocentrics (Chromosomes 7, 8, 9, 10, 11, and 12) in diploid L. tigrinum, two pairs of metacentrics (Chromosomes 1 and 2), six pairs of subtelocentrics (Chromosomes 3, 4, 5, 6, 7, and 11) and four pairs of telocentrics (Chromosomes 8, 9, 10, and 12) in triploid L. tigrinum. Ten (Chromosomes 1, 2, 6, 7, and 11) or twelve (Chromosomes 1, 2, 4, 6, 7, and 11) loci of 45S rDNA were detected in the diploid chromosomes whereas fifteen loci were detected in triploid chromosomes (Chromosomes 1, 2, 6, 7, and 11). Two loci of 5S rDNAs were located close to each other at the both diploid and triploid chromosome 3. New cytotypes of secondary constriction were observed in diploid (1-1-1-0) and triploid (3-2-1-1).

Fifty-two populations representing thirty-two taxa of Lilium L. from China are karyologically analyzed. The results showed that all populations have the same basic chromosome number x012, and all species are diploid except Lilium tigrinum, which is triploid with 2n036. In addition, one population of L. sulphureum is aneuploid with 2n023. The karyotype evolution in the genus Lilium is mainly in terms of alterations of the fine structure of the chromosomes, not in ploidy or basic chromosome number variation. When combining previous studies and our results, we consider the major driving forces for evolution of Lilium to be an increase in karyotypic asymmetry, and unequal translocation and formation of secondary constrictions. The existence of intercalary satellites on the two largest pairs of chromosomes is correlated with the geographic distribution pattern of Lilium across the world. The Hengduan Mountains is a modern diversity and differentiation center of Lilium and it also could be the center of origin for this genus. In addition, some taxonomic conclusions were verified on the subgenus level. Among the species investigated, the chromosome number and karyotypes of 13 taxa were documented for the first time.
