Genetic resources of red clover (Trifolium pratense L.) are the basis for the improvement of this important forage legume. The objective of this study was microsatellite characterization of the accessions from the collection of the Institute of Field and Vegetable Crops in Novi Sad, Serbia. Molecular evaluation of 46 red clover genotypes was performed by applying the set of 14 primer pairs of microsatellite markers. These primer pairs amplified a total of 187 alleles, with an average of 13.36 alleles per locus and average polymorphism information content (PIC) value was 0.306. The minimum values of Dice genetic distances based on polymorphism of microsatellite markers were found among genotypes NCPGRU2 and NCPGRU5 (0.311) and the highest values of genetic distances were determined for a couple of genotypes Violeta and BGR2 (0.933). The average genetic distance between all pairs of genotypes amounted 0.587. The results of the principal coordinate analysis (PCoA) were consistent with the results obtained on the basis of cluster analysis, except that the PCoA allocated another four genotypes. There was no relationship between groups of genotypes formed by the use of cluster analyses and PCoA with their geographical origin. Analysis of molecular variance of 46 red clover genotypes by the status and ploidy level was significant, but it also suggested a weak genetic differentiation of groups formed on the basis of those characteristics. Observed groups of genotypes, according to the cluster analyses and PCoA of microsatellite data, could be used in future breeding programs for the selection of germplasm.

This research comprises a karyologic study of the nine taxa of Trifolium from Turkey. From the Trifolium L. section, Trifolium striatum L. and T. leucanthum Bieb. chromosome numbers were 2n = 14, T. echinatum Bieb. was 2n = 16 and T. phleoides Pourr. ex Willd was 2n = 12. From the Vesicaria Crantz. section, T. resupinatum L. var. resupinatum, T. clusii Godr. and Gren., and T. tomentosum L. all had chromosome numbers of 2n = 16. Chromosome numbers of T. retusum, L. from section Lotoidea, Crantz. and T. glanduliferum Boiss. var. glanduliferum from section Paramesus (C. Presl) Gris. were all found to be 2n = 16. Seven of the 9 taxa contained all pairs of chromosomes and were metacentric; the remaining taxa included 7 were metacentric and only one pair was submetacentric. One pair of satellites was found in T. phleoides. This study reports detailed karyotype analyses from Turkey for the first time in Trifolium.

DNA barcoding is an important molecular approach in the determination of species diversity, evaluation of phylogenetic relationships and identification of taxonomically problematic species because of insufficient diagnostic characters. It has great importance to determine the barcoding regions that will give the best result in the evaluation of phylogenetic relationships in species like Trifolium that spread over wide geographical regions in the world and show high variation accordingly. For this aim, matK and rbcL regions belonging to cpDNA was used to determine the barcoding ability and evaluate the taxonomy of the genus Trifolium. It is observed that both of barcoding regions are valuable to reveal the phylogenetic relationships, in addition to their grouping ability the species as taxonomically. However, especially the using of matK sequence informations that have high variable sites (158) and grouping ability clearly for all taxa is strongly recommended. Results provided from this study support the phylogenetic classification suggested by Ellison et al. (2006).
