Isolation of Adipose-Derived Stem/Stromal Cells from Cryopreserved Fat Tissue and Transplantation into Rats with Spinal Cord Injury

Abstract: Adipose tissue contains multipotent cells known as adipose-derived stem/stromal cells (ASCs), which have therapeutic potential for various diseases. Although the demand for adipose tissue for research use remains high, no adipose tissue bank exists. In this study, we attempted to isolate ASCs from cryopreserved adipose tissue with the aim of developing a banking system. ASCs were isolated from fresh and cryopreserved adipose tissue of rats and compared for proliferation (doubling time), differentiation capabil… Show more

“…As adherent cells reached 80% confluence (passage 1: P1), hADSCs were detached with 0.25% trypsin/1mM ethylenediamine tetraacetic acid (EDTA) (Sigma) and plated in flasks at a density of 1,5×10 4 cells/75 cm 2 (passage 2: P2). The cellular density was determined by manually counting the number of cells at each division 36 . Identified as cell 1 (C1). Patient-derived hAT-MSCs: cells were obtained from the subcutaneous fat of a 30-year-old female, submitted to abdominal liposuction at the Hospital de Clínicas in Porto Alegre, Brazil.…”

“…The cellular density was determined by manually counting the number of cells at each division 36 Both cells C1 and C2 were expanded under the same conditions and used only from 4 th to 8 th passage 26,34,37 . An aliquot of 1x10 4 hAT-MSCs was placed on a slide and analyzed by immunofluorescence.…”
