Isolation and molecular characterization of a new Muscovy duck parvovirus from Muscovy ducks in the USA

Abstract: Between 1997 and 1999 several cases of a new disease in Muscovy ducks were reported in Pennsylvania, USA. The cases were characterized by locomotor dysfunction, weakness, recumbency, 40 to 60% morbidity and 10 to 40% mortality. The most characteristic microscopic lesions were moderate to severe degenerative rhabodomyopathy. In order to characterize the aetiological agent, virus isolation was attempted from the spleen, liver, heart, skeletal muscle and intestine by inoculation of 14-day-old Muscovy duck embryos… Show more

“…Poonia B reported new duck parvovirus from Muscovy duck, only shared 84.6% sequence identity with GPV and 84.5% identity with MDPV [8] . Wang reported a part of VP gene sequence of the GPV PT strain isolated from Muscovy ducks had the characteristics of MDPV sequence [9] .…”

“…The two viruses possess approximately 85% homology in their overall protein sequences and have been shown to be antigenically related to each other. Nonetheless, GPV and MDPV are distinct, and as a result, any antibody-driven cross-protection between these two closely related viruses is largely limited (5,6).…”

“…It is almost impossible to find an MDPV antibody-free egg, as the appropriate detection test is not available. However, it should be possible to cultivate MDPV in MDPV antibody-free Muscovy duck eggs [1].…”

“…The Avian Influenza H5N1 detection was conducted as previously published [21]. The presence of MDPV was confirmed using PCR with primer pairs covering the NS1 [12] or REP [6], hereinafter is mentioned as NS1-REP, and VP1 genes as previously described [1] and then sequencing the results at the Eijkman Institute Jakarta using BigDye® Terminator v3.1 Cycle Sequencing Kit (Life Technologies) following the accompanying manual. The total DNA was isolated and purified from heart and liver tissue homogenates, as well as from the allantois fluid of inoculated Muscovy duck eggs, using a DNA isolation kit (Invitrogen) following the protocol in the provided manual.…”
