Isolation and Identification of Endophytic Fungi from Leave and Stem of Calopogonium mucunoides

Abstract: Thirty-seven isolates of endophytic fungi were isolated from leaves and stems of Calopogonium mucunoides collected from PTPN PTPN XII (Persero) Rubber Plantation, Klatakan, Kecamatan Tanggul, Kabupaten Jember, Jawa Timur. All isolates were identified based on morphological characteristics using the light microscope. The 37 isolates of endophytic fungi are members of Deuteromycota and Basidiomycota and classified to genera Phoma, Phomopsis, Corynespora, Rhizoctonia, Helicosporium, Curvularia, Torulomyces, Glioc… Show more

“…was from the stems. There are factors determined the presence of endophytic fungi in plants, such as environment, species, and isolation technique [11]. Figures 1 and 2 display the macroscopic and microscopic morphologies of identified endophytic fungi that were observed and recorded based on the Illustrated Genera of Imperfect Fungi Handbook [10].…”

“…Endophytic fungus isolated from the leaves, stem bark, and root bark of S. malaccense plants from Palembang, South Sumatra, produced various antibacterial substances depending on their taxonomic groupings. These three plant parts could have varied functions in supporting host growth and nutritional needs in pathogen protection (Selim et al 2012;Fitriarni and Kasiamdari 2018). Endophytic fungi create secondary compounds that protect host plants against pathogens, consequently, plant parts can produce a variety of endophytic fungal isolates (Fadiji and Babalola 2020).…”

“…Purification was carried out by transferring the colonies to a new PDA medium with a single spore isolation method, then incubated at room temperature for 2 x 24 hours. The purified fungal colonies were re-cultured on PDA media as working culture (in Petri dishes) and stock cultures (in test tubes) (Fitriarni and Kasiamdari 2018;Hanin and Fitriasari 2019;Habisukan et al 2021).…”
