SUMMARY: The Mexican rockfish Sebastes macdonaldi is the Northeast Pacific rockfish with the southernmost distribution, featuring isolated populations in the Gulf of California. We analysed seven microsatellite loci in 111 organisms collected throughout most of its geographical range to test long-standing hypotheses regarding its disjunct distribution. One locus was fixed and the number of alleles in polymorphic loci ranged from 2 to 24 (average 13.5). We found very high levels of polymorphism (overall He = 0.75) comparable to other congeneric species but no significant differences in genetic diversity among localities or between Pacific and Gulf of California populations (p > 0.1). Significant shifts in allelic and genotypic frequencies were detected at three loci, which resulted in a small but significant partitioning of genetic variance among California, Baja California and Gulf of California populations (F ST = 0.007, p = 0.03) and between Gulf and Pacific populations (F ST = 0.01, p = 0.004). The latter but not the former result was corroborated by analyses of molecular variance (AMOVA) using the number of distinct alleles (F ST ) and the sum of square differences of allele sizes (R ST ) as Euclidean distances. The evidence argues against contemporary gene flow between the gulf and the Pacific ocean and against an ancient invasion of the gulf with a founder effect. The small level of divergence favours a recent dispersal but a larger data set including DNA sequences amenable to phylogenetic analyses will help to test alternative hypotheses of dispersal versus vicariance.Key words: Mexican rockfish, disjunct distribution, microsatellites, genetic structure, Baja California, México. ABSTRACT. The population structure of the black rockfish, Sebastes inermis (Sebastidae), was estimated using 10 microsatellite loci developed for S. schlegeli on samples of 174 individuals collected from three wild and three hatchery populations in Korea. Reduced genetic variation was detected in hatchery strains [overall number of alleles (N A ) = 8.07; allelic richness (A R ) = 7.37; observed heterozygosity (H O ) = 0.641] compared with the wild samples (overall N A = 8.43; A R = 7.83; H O = 0.670), but the difference was not significant. Genetic differentiation among the populations was significant (overall F ST = 0.0237, P < 0.05). Pairwise F ST tests, neighbor-joining tree, and Population genetic structure of black rockfish in Korea principal component analyses showed significant genetic heterogeneity among the hatchery strains and between wild and hatchery strains, but not among the wild populations, indicating high levels of gene flow along the southern coast of Korea, even though the black rockfish is a benthic, non-migratory marine species. Genetic differentiation among the hatchery strains could reflect genetic drift due to intensive breeding practices. Thus, in the interests of optimal resource management, genetic variation should be monitored and inbreeding controlled within stocks in commercial breeding programs. Information on genetic population structure based on cross-species microsatellite markers can aid in the proper management of S. inermis populations.

Rockfish species of the genus Sebastes are notable for being numerous and diverse. Rockfishes are unusual among fish because they fertilize their eggs internally and release live, swimming larvae. They undergo complex courting behaviors, which may allow females to be selective about their mates. The major histocompatibility complex (MHC) is implicated as having an important influence on mate selection in other fishes, especially in sticklebacks and salmonids. Research suggests that females choose mates that optimize the MHC genotypes of their offspring. Previous research on rockfishes indicates that multiple functional MHC sequences may be found in each species, and that multiple mating is common in the genus, possibly as a bet-hedging strategy against uncertain or incomplete mate-selection information. In this project, we characterized the MHC genotypes of copper (S. caurinus) and quillback (S. maliger) rockfish parents, assessed parentage of 14 larval broods, and assessed the MHC genotypes of the parents to determine if MHC-mediated mate choice was occurring. As in previous studies, we found that rockfishes possess multiple, highly variable MHC genes, and that females may mate with multiple males. We also found evidence of female preference for particular males. However, we found no strong evidence of selection based on MHC genotype. Females were not consistently selective based on relatedness, allele count, proportion of shared alleles, or minimum, mean, or maximum DNA or amino acid genetic distance. Instead, it appears that females were selective based on other measures of mate quality not considered in this study, with some hedging of bets through multiple mating also occurring.
