The objective of this work was to investigate the effects of the methanol extracts of Gentiana cruciata L. aerial parts (GCA) and roots (GCR) against carbon tetrachloride-induced liver injury in rats. Pretreatment with GCA and GCR, containing sweroside, swertiamarin and gentiopicrin in high concentrations, dose-dependently and significantly decreased the levels of serum transaminases, alkaline phosphatase and total bilirubin, whereas an increase in the level of total protein was found compared with the CCl4-treated group. Moreover, oral administration of extracts significantly enhanced antioxidant enzyme activities (superoxide dismutase and catalase), increased the content of glutathione and decreased the content of TBARS. Microscopic evaluations of the liver revealed CCl4-induced lesions and related toxic manifestations that were minimal in the liver of rats pretreated with extracts at the dose of 400 mg per kg b.w. The results suggest that the use of G. cruciata extracts has a merit as a potent candidate in protecting the liver against chemical induced toxicity.

Die Rolle des Arbeitsgruppenleiters eines chirurgischen Forschungslabors und seine Umgebung -Eine persönliche SichtZusammenfassung. Grundlagen: Im vorliegenden Manuskript werden die Anforderungen und Aufgaben des Laborleiters eines chirurgischen Forschungslabors ebenso diskutiert, wie die Umgebung des Labors, und Folgen neuer universitärer Karrieremodelle für die chirurgische Forschung.Methodik: Diese Arbeit basiert auf persönlichen Erfahrungen und Ansichten.Ergebnisse: Der Leiter eines chirurgischen Forschungslabors -ob Mediziner oder Naturwissenschaftler -hat zahlreiche Aufgaben zu bewältigen, die sich von der Motivation von Mitarbeitern, und kommunikativen Aufgaben ü ber das Selektionieren von relevanten Informationen zwischen Klinik und Labor und der wissenschaftlichen Qualität von Projekten, bis hin zur Ü bersetzung von Labordaten in klinische Studien spannen.Schlussfolgerungen: Nachdem ein Laborleiter nur so gut sein kann wie sein Team und seine Kooperationspartner -inklusive Chirurgen die sich für die Forschung interessieren -ist die Bildung einer effizienten Arbeitsgruppe mit einer kritischen Masse sicher eine seiner Hauptaufgaben. Background: This manuscript summarises the tasks and skills of a laboratory head in surgical research, describes the laboratory environment and discusses the consequences of novel career models for clinicians on surgical research.Methods: This work is based on personal experiences and views.Results: The laboratory head has to accomplish several tasks ranging from communication, motivation of co-workers, filtering of information and scientific quality of projects, to transferring laboratory research data to studies in humans.Discussion: A laboratory head can be only be as good as his team and co-operations partners, therefore the setting up of an efficient team of a critical mass, including surgeons interested in research, is a central element in the laboratory heads' tasks. Scientific quality must be assured and the potential significance of projects has to be evaluated to support and advance the most promising attempts in a scientifically correct and efficient way.

A novel, fast, and simple capillary electrophoresis method has been developed for the analysis of gentisin, isogentisin, and amarogentin in roots of Gentiana lutea (yellow gentian), an herb traditionally used as gastric stimulant. Gentisin and isogentisin are xanthones showing potent inhibition of monoamine oxidase type A and B, amarogentin represents one of the bitter principles of Gentiana, responsible for its gastric-roborant effects. Optimal CE-separation conditions comprise a 100 mM sodium tetraborate buffer of pH 9.3, containing 10 mM beta-cyclodextrin as additive; optimum temperature and applied voltage were found to be 30 degrees C and 25 kV, respectively. Direct diode array detection at 260 nm (gentisin, isogentisin) and 242 nm (amarogentin) was performed, and the required analysis time was only 11 min. The developed method was validated for linearity, sensitivity, precision, and accuracy, and utilized to assay several commercially available G. lutea samples. Quantitative data obtained with the developed CE method are compared with HPLC results, and the advantages of each approach are discussed.
