Galium verum, also known as Lady's Bedstraw, is a herbaceous perennial plant of the family Rubiaceae, native to Europe and Asia and used in traditional medicine as an anticancer medicine. It is used as a decoction in most traditional recipes, applied externally as well as internally. We produced a Galium verum decoction and applied it in vitro to chemosensitive (Hep-2 and HLaC79) and chemoresistant, P-glycoprotein-overexpressing (Hep2-Tax, HLaC79-Tax) laryngeal carcinoma cell lines. It could be demonstrated that Galium aqueous extract is cytotoxic for all cell lines. A detailed spheroid-based 3D invasion analysis of Hep2 and Hep2-Tax in semisolid collagen gels and on different extracellular matrix coatings was performed, which showed an inhibition of invasion by sublethal concentrations of Galium decoction and proved to be even more pronounced in the more aggressively invading chemoresistant Hep2-Tax cell line. Gelatinolytic activity of MMP-2 was downregulated in three of the four cell lines. Angiogenesis (endothelial tube formation) in contrast, was not affected by Galium aqueous extract. Gene expression array on HLaC79 and Hep2 cell lines treated with Galium decoction vs. untreated controls revealed no unique pathway activation patterns in these cells. Results are discussed with respect to the use of herbal drugs as a preventive and/or a concomitant therapeutic approach in head and neck cancer.

Abstract:The antioxidant properties of methanol extracts of Lady's Bedstraw (Galium verum L., Rubiaceae) herb from two different localities in Serbia were evaluated. Antioxidant activity was assessed in four different model systems. Free radical scavenging capacity (RSC) was examined by measuring the scavenging activity of extracts on 2,2-diphenyl-1-pycrylhydrazil (DPPH) and hydroxyl radical (OH), as well as on hydrogen peroxide. In addition, the protective effects of lipid peroxidation (LP) in corn oil were evaluated by the TBA-assay using the Fe 2+ /ascorbate system of induction. The amount of dried extract, the content of total phenolics, flavonoids and chlorophylls was also determined. Extracts from both locations expressed very strong scavenger activity, reducing the DPPH• (IC 50 =3.10 μg/ml and 8.04 μg/ml) and OH radical formation (IC 50 =0.05 μg/ml and 0.54 μg/ml) and neutralising H 2 O 2 (IC 50 =4.98 μg/ml and 3.80 μg/ml), in a dose dependant manner. Also, examined extracts showed notable inhibition of LP (IC 50 =11.69 μg/ml and 19.47 μg/ml). The observed differences in antioxidant activity could be partially explained by the levels of phenolics (2.44-4.65 mg and 4.57-5.16 mg gallic acid equivalents/g dry extract), flavonoids (6.38-10.70 μg and 15.56-17.96 μg quercetin equivalents/g dry extract) and chlorophylls in the investigated Lady's Bedstraw extracts. © Versita Sp. z o.o.

“…Recently, we studied the constituents of this plant and provided the characterization of a new acyl flavonol diglycoside, kaempferol 3-O-[2¢¢-O-(E-6¢¢¢-O-feruloyl)-b-D-glucopyranosyl]-b-D-galactopyranoside (9), together with quercetin, quercetin 3-O-glucopyranoside, quercetin 3-O-sambubioside (7), and quercetin 3-O-sophoroside (8). In addition, some known compounds including b-sitosterol, b-sitosteryl glucoside, iridoid V 3 (1), an artificial product of asperuloside (2) (3), asperuloside (2), asperulosidic acid methyl ester (3), (E)-6-O-p-methoxycinnamoylscandoside methyl ester (4), (E)-6-O-feruloylscandoside methyl ester (5) and (E)-6-Ocoumaroylscandoside methyl ester (6) (4 ± 6) were also obtained. Superoxide (´O 2 ± ) and other reactive oxygen species (ROS) are involved in many pathological processes such as some inflammatory diseases, hepatitis, ageing, cancer, etc (7).…”

A study on the bioactive principles of Hedyotis diffusa Willd., led to the isolation of a new acyl flavonol di-glycoside which was characterized as kaempferol 3-O[2"-O-(E-6'"-O-feruloyl)-beta-D-glucopyranosyl]-beta-D-galactop yranoside by spectral and chemical methods from the methanolic extract. In addition, three known flavonol glycosides and six known iridoid glycosides were also obtained. The above-mentioned glycosides were tested for antioxidant effects on xanthine oxidase inhibition, xanthine-xanthine oxidase cytochrome c and TBA-MDA systems.
