Iodine Conjugated Pt(IV) Nanoparticles for Precise Chemotherapy with Iodine–Pt Guided Computed Tomography Imaging and Biotin-Mediated Tumor-Targeting

Abstract: Theranostics of platinum (Pt)-based chemotherapy are able to self-track the biodistribution and pharmacokinetics while performing therapeutic effects. Pt-based CT imaging is expected to visualize and monitor the tumor throughout the entire tumor inhibition stage. However, a sufficient Pt concentration is necessary for CT imaging, which may bring about severe nephrotoxicity. A Bio-Pt-I compound is designed and synthesized by conjugation of iodine and biotin to the structure of Pt and further self-assembles into… Show more

“…Although with many successes, the generation of most of these polymer microcarriers often requires additional extra initiators. In contrast, the photo-activatable platinum(IV)-azide complexes (Pt(IV) prodrugs) have been previously encapsulated in nano/microcarriers for tumor therapy due to its transformation to highly cytotoxic Pt(II) species [ [37] , [38] , [39] , [40] , [41] ]. During this photo-activation process, the N 3 • could also be generated from Pt(IV).…”

“…However, high concentrations of Pt must be maintained to obtain satisfactory CT imaging results, raising the possibility of serious side effects 139 . Recently, Huang et al synthesized reductant-sensitive bio-Pt-I NPs consisting of iodine-conjugated Pt (IV) compounds and biotin as a theranostic nanomedicine for CT image-guided treatment ( Figure 8 I ) 140 . The bio-Pt-I NPs with high Pt (25.36%) and I (32.99%) contents could be used for CT imaging and real-time dynamic distribution monitoring of nanomedicines.…”

“…22,23 Recently, iodine nanoparticles were found to be effective in enhancing proapoptotic genes, and these iodine-based materials are also applicable in tumour tissue diagnosis using the computed tomography technique. 24 So, here, we hypothesise that forming composite particles consisting of copper and iodine will provide a synergistic effect against microbial and cancer cell growth, as iodine will enhance the antibacterial effect of copper. At the same time, copper will enhance the anticancer effect of iodine.…”
