Involvement of p38 MAPK in Leydig cell aging and age-related decline in testosterone

Abstract: IntroductionAge-related decline in testosterone is associated with Leydig cell aging with impaired testosterone synthesis in aging. Obesity accelerates the age-related decline in testosterone. However, the mechanisms underlying the Leydig cell aging and the effects of obesity on Leydig cell aging remain unclear.MethodNatural aging mice and diet-induced obese mice were used to assess the process of testicular Leydig cell senescence with age or obesity. Bioinformatic analysis of the young and aged human testes w… Show more

“…The ligandreceptor pairs between Leydig cells and other germ cell were significantly reduced in older samples. In line with that, multiple studies also reported that Leydig cells from older men display malfunction, including lower production of testosterone [13,26]. So, application of TRT to treat male infertility was mentioned [27].…”

“…In this study, FGFR3 and TGFβ2 differentially expressed genes were signi cantly enriched in the MAPK signaling pathway in testis tissues. Several other studies [41][42][43] have shown that this pathway is involved in cell proliferation, differentiation, and apoptosis, making it an important signaling pathway. In mammals, MAPK can be translated as "mitogen-activated protein kinase" Regulating cell proliferation, differentiation, movement, and survival is one of the important pathways affecting testis development and spermatogenesis [44].…”

“…LPO, AGEs, and resulting ROS can directly interfere with mitochondrial DNA and RNA replication, resulting in mitochondrial structure damage and dysfunction, which in turn leads to more ROS production 30 . In the testis, Leydig cells are prime targets for macromolecular OS and cell aging, which results in testosterone synthesis reduction 33 . Antioxidant therapy is effective in the treatment of hyperglycemia‐related male reproductive dysfunction.…”
