Intra- and inter-individual comparison of Porphyromonas gingivalis genotypes

Abstract: Genetic analysis of 31 clinical strains of Porphyromonas gingivalis isolated from nine subjects, 2-6 strains per subject, was performed by Southern hybridization. Chromosomal DNA was extracted by the method of Moncla et al. [1] and digested to completion with restriction endonucleases PstI, ClaI and BglI. The DNA fragments were separated electrophoretically on agarose gels, transferred to nylon membranes and hybridized to the non-radioactively labelled plasmid pKK 3535 which contains the rmB ribosomal RNA oper… Show more

“…III (VMGA III) medium, and cultured. A. actinomycetemcomitans and P. gingivalis were identified as previously described (18, 19). Briefly, for A. actinomycetemcomitans , 100-µl aliquots of undiluted and 10 −2 dilutions of samples were inoculated on Trypic soy bacitracin vancomycin (TSBV)-agar plates and incubated in 5% CO 2 in air at 36°C for three days.…”

“…Caucasian subjects usually harbour one P . gingivalis clonal type (Loos et al 1992, Saarela et al 1993a, 1996, van Steenbergen et al 1993) but multiple clones per subject have also been reported (Enersen et al 2006). Recently, van Winkelhoff et al (2007) reported on multiple clonal types in 34.5% of the subjects in an Indonesian population with varying degrees of periodontal disease.…”

“…Socransky and Martin [34] and Saarela et al [35] used ribotyping to distinguish clonal types of P. gingivalis. In this method, chromosomal DNA is digested with restriction endonucleases, subjected to electrophoresis and Southern blotting, after which it is hybridised with ribosomal DNA.…”

“…In this method, chromosomal DNA is digested with restriction endonucleases, subjected to electrophoresis and Southern blotting, after which it is hybridised with ribosomal DNA. Isolates from six [34] and nine [35] different subjects, respectively, were found to be of different types. An advantage of this method compared with whole genomic DNA fingerprinting is the lower number of bands obtained, which facilitates comparison of lanes.…”
