Integration of pharmacokinetic–pharmacodynamic for dose optimization of doxycycline against Haemophilus parasuis in pigs

Abstract: The aims of this study were to establish optimal doses of doxycycline (dox) against Haemophilus parasuis on the basis of pharmacokinetic-pharmacodynamic (PK-PD) integration modeling. The infected model was established by intranasal inoculation of organism in pigs and confirmed by clinical signs, blood biochemistry, and microscopic examinations. The recommended dose (20 mg/kg b.w.) was administered in pigs through intramuscular routes for PK studies. The area under the concentration 0- to 24-hr curve (AUC ), el… Show more

“…For bacteria with the MIC value of 0.4 and 1.5 µg/mL, the T>MIC ratios of doxycycline administered at a dose of 20 mg/kg and a dose interval of 24 h were 152% and 54%, respectively. The AUC 0–24 /MIC ratios to achieve the bacteriostatic and bactericidal effect of doxycycline against Haemophilus parasuis in pigs have been determined as 59 and 98, respectively [ 32 ]. In the present study, an AUC 0–24 /MIC ratio of 100.75 for bacteria with the MIC value of 0.4 µg/mL was higher than that previously reported for optimum bacteriostatic and bactericidal activity [ 32 ].…”

In goats, a 20 mg/kg oral dose of doxyxycline can provide the efficient treatment of infections caused by susceptible bacteria with the MIC value of ≤0.4 µg/mL.…”

“…This PK/PD model reflects the relationship between the drug, bacteria, and quantification of the activity (as well as the likely efficacy) of antimicrobial agents against target pathogens (Zhang et al, 2018a). The dynamic model that we used provided a continuous flow of doxycycline to simulate the PK of doxycycline in pigs.…”

“…Numerous PK studies of DC have been conducted in chickens (6), ducks (7), pigs (8), sheep (9), and goats (10). Few studies have been implemented in fish, such as tilapia (11) and grass carp (12,13).…”
