“…From a chemical point of view, drugs are incorporated within hydrogels by two general modalities, through either chemical or physical interactions. In a recent work, Zhang et al developed a pH-responsive and injectable peptide hydrogel–MTX delivery system for local and specific tumor chemotherapy, which afforded low side effects [ 76 ]. A hydrophilic drug, methotrexate (MTX) was conjugated to the peptide chain (KKFKFEFEF) using a pH-responsive linker, 2,3-dimethylmaleic anhydride, via the formation of amidic bonds.…”

“…This approach has been recently widely applied in the field of pH-responsive self-assembling peptide-based biomaterials, as the nonfunctional molecule becomes functional when the pH changes, providing a specific and “smart” target treatment. Various pH-labile linkages such as hydrazones, cis -aconityls, dimethylmaleic amides, and acetals (Figure B) are commonly used in pH-responsive systems. For example, Balci and Top designed a pH-responsive drug delivery system by conjugating mPEG-peptide with Dox via an acid cleavable hydrazone bond to evaluate drug release under normal and acidic conditions .…”

“…Zhang et al developed an injectable pH-responsive drug–peptide hydrogel for local chemotherapy [ 52 ]. Firstly, methotrexate (MTX), a small-molecule drug with carboxyl groups and 2,3-dimethylmaleic anhydride (DA), a pH-responsive linker, were directly conjugated onto the KKFKFEFEF peptide chain via the amidation reaction, achieving a drug-loaded pH-responsive peptide composite (MTX-KKFKFEFEF(DA)).…”
