Inhibitory effects of the ethyl acetate extract from bulbs ofScilla scilloideson lipoxygenase and hyaluronidase activities

Abstract: Context: Scilla scilloides Druce (Liliaceae) is a folk medicine to treat dermal inflammation; however, the medicinal properties of this plant have not been completely established. Objective: The current study investigates the potent anti-inflammatory effects of S. scilloides bulbs for its traditional usage using lipoxygenase and hyaluronidase as the inflammation model. To gain insight into the active constituents, nine homoisoflavones (1-9) were subsequently tested. Materials and methods: Lipoxygenase and hyal… Show more

“…This study employed tannic acid as a positive control for comparison purposes. Figure 6 reveals that tannic acid had a higher hyaluronidase activity inhibition; the IC50 was 0.14 mg/mL, similar to the value obtained by Nishida et al (0.121 mg/mL; 71.1 mM) [42]. By contrast, an IC50 of 8.91 mg/mL was calculated for the RPH solution.…”

“…Therefore, enzymes control the size and concentration of depolymerized hyaluronan chains. Depolymerization can begin pathological processes, for example, increasing endothelial permeability or remodeling tissue during allergic reactions [32][33][34]. The non-steroidal anti-inflammatory drugs (NSAIDs) are commonly used to treat inflammation, but due to their side effects, their prolonged intake should be limited.…”

“…Hyaluronidase inhibitory assay-The hyaluronidase activity was measured using the Morgan-Elson reaction with minor modifications [45][46][47][48]. In a total volume of 175 μL, the assayed inhibitors (50 μL in 9-14% DMSO) were mixed with 25 μL of testes bovine hyaluronidase (0.3 mg mL −1 of 0.1 M sodium acetate buffer with 3.5 mM CaCl 2 and 8.5 mg mL −1 NaCl, pH 5.2) and incubated at 37°C for 20 Min.…”
