SummaryBackground: equine gastric ulcer syndrome (EGUS) has a multifactorial nature and it affects both the squamous and glandular mucosa of the stomach. Multiple therapeutic strategies involving large periods of medication are used to treat this condition. Objective: to evaluate the effects of administering corn oil (CO) to horses with induced gastric ulcers, and to describe the mechanism of action of the mucosal repair. Methods: fifteen horses divided into three groups were used. A combination of confinement and phenylbutazone was used during the ulcer-induction phase. After the gastroscopic evaluation and laboratory tests, animals were treated with sucralfate (SA; group I), and CO at doses of 70 and 90 mL/100 Kg (groups II and III, respectively). Gastroscopy, gastric content collection, and biopsy of the glandular mucosa were performed to analyze prostaglandin E 2 (PGE 2 ), pH, and antioxidant and oxidant parameters. Results: mild to moderate gastric lesions were induced in the glandular and non-glandular mucosa. Among the mechanisms of the treatments, reestablishment of the antioxidant parameters and inhibition of myeloperoxidase (MPO) oxidant enzyme were prominent, but PGE 2 concentration had a weak influence. Conclusion: similarly to SA, CO only had therapeutic effects in the glandular mucosa.

Synthetic matrices have been used widely to repair and/or to replace biological tissues. However, there is relatively little information on the effect of different anatomical compartments on the host response to foreign implants. We have analyzed such responses to sponge implants in subcutaneous and in intraperitoneal sites in mice at days 3, 5, and 8 postimplantation by measuring inflammation, angiogenesis, and production of proangiogenic/inflammatory cytokines. The angiogenic response, assessed by hemoglobin content and by morphometric analysis of the number of vessels, was higher in intraperitoneal implants. Levels of vascular endothelial growth factor in intraperitoneal implants were 14-fold higher than in subcutaneous implants at day 3 and remained high for the next 5 days. Neutrophil accumulation as determined by myeloperoxidase activity was the same in both types of implants. Macrophage accumulation (N-acetylglucosaminidase activity) was also similar on days 3 and 8 in both implants. Levels of the chemokine CXCL2/KC were always higher, but those of CCL2/JE lower, in the intraperitoneal implant. These results demonstrate that the anatomical site of the implant markedly influenced the host response to synthetic matrices. Our results provide a greater understanding of factors affecting the biocompatibility of exogenous materials placed at different anatomical sites.

The 15-deoxy-⌬ 12,14 -prostaglandin J 2 (15d-PGJ 2 ) is an endogenous ligand of peroxisome proliferator-activated receptors ␥ (PPAR-␥) and is now recognized as a potent anti-inflammatory mediator. However, information regarding the influence of 15d-PGJ 2 on inflammatory pain is still unknown. In this study, we evaluated the effect of 15d-PGJ 2 upon inflammatory hypernociception and the mechanisms involved in this effect. We observed that intraplantar administration of 15d-PGJ 2 (30 -300 ng/paw) inhibits the mechanical hypernociception induced by both carrageenan (100 g/paw) and the directly acting hypernociceptive mediator, prostaglandin E 2 (PGE 2 ). Moreover, 15d-PGJ 2 [100 ng/temporomandibular joint (TMJ)] inhibits formalininduced TMJ hypernociception. On the other hand, the direct administration of 15d-PGJ 2 into the dorsal root ganglion was ineffective in blocking PGE 2 -induced hypernociception. In addition, the 15d-PGJ 2 antinociceptive effect was enhanced by the increase of macrophage population in paw tissue due to local injection of thioglycollate, suggesting the involvement of these cells on the 15d-PGJ 2 -antinociceptive effect. Moreover, the antinociceptive effect of 15d-PGJ 2 was also blocked by naloxone and by the PPAR-␥ antagonist 2-chloro-5-nitro-Nphenylbenzamide (GW9662), suggesting the involvement of peripheral opioids and PPAR-␥ receptor in the process. Similar to opioids, the 15d-PGJ 2 antinociceptive action depends on the nitric oxide/cGMP/protein kinase G (PKG)/K ATP ϩ channel pathway because it was prevented by the pretreatment with the inhibitors of nitric-oxide synthase (N G -monomethyl-L-arginine acetate), guanylate cyclase ]1H-(1,2,4)-oxadiazolo(4,2-␣)quinoxalin-1-one [, PKG [indolo[2,pyrrolo [3,4-c]carbazole aglycone (KT5823)], or with the ATP-sensitive potassium channel blocker glibenclamide. Taken together, these results demonstrate for the first time that 15d-PGJ 2 inhibits inflammatory hypernociception via PPAR-␥ activation. This effect seems to be dependent on endogenous opioids and local macrophages.
