The present study evaluated the impact that orchiectomy, a routine but painful intervention in bovine husbandry, can cause on pulmonary immunity. To identify whether orchiectomy can impair lung defense, analyses of serum cortisol concentration and of alveolar macrophage and their function (phagocytosis and respiratory burst) were evaluated. Sixteen Holstein bulls (7 mo old, 250±50kg of body weight BW) were divided in two homogeneous groups -the castrated group and the sham group -and the sample were collected on Days -7, 1 and 7 relative to the day of the procedure. Serum cortisol concentration statistically increased on Days 1 and 7 (D-7: 4,97±1,28ng/ml; D1: 6,35 ±1,10ng/ml; D7: 8,28±1,94ng/ml) after castration and these variables seem to impact the alveolar macrophage percentage on D1 (D-7: 76,86±3,44%; D1: 60,92±2,44%; D7: 74,17±2,56%) and their respective function of phagocytosis (P) and the oxidative burst (OB) (D-7: 4,97±1,28ng/mL; D1: 6,35 ±1,10ng/mL; D7: 8,28±1,94ng/mL). Bras. 31(3): 244-246, março 2011 RESUMO.-Com o intuito de verificar a influência de diferentes condições meteorológicas na sanidade de bovinos, foi realizado citologia de lavados traqueobrônquicos obtidos por traqueocentese e leucograma sanguíneo de cinco bezerros em situações de extremos de temperatura ambiental, sendo T1 = T (temperatura ambiental) de 5 o C e UR (umidade relativa do ar) 93%; T2 = temperatura controle de T 22 o C e UR 80%; e T3 = T 30 o C e UR 41%. Pode-se observar que a condição T3 provavelmente gerou estresse nos animais, pois se observou monocitose significativa no leucograma e na análise do lavado traqueobrônquico, uma diminuição significativa de macrófagos alveolares gigantes, provavelmente por diminuição da atividade macrofágica alveolar, caracterizando esta temperatura e umidade relativa do ar como favorá-veis ao aparecimento de doenças respiratórias.TERMOS DE INDEXAÇÃO: Aparelho respiratório, temperatura ambiental, bovinos. INTRODUÇÃODiariamente os bovinos são expostos a diversos fatores agressores, e mesmo assim a maioria deles se mantém saudáveis devido a eficientes mecanismos de defesa física, celular e humoral (Liggitt 1985). No entanto situações como extremos de temperatura, transporte, união de animais de várias origens são causas comuns de estresse em bovinos que podem provocar alterações fisiológicas e hematológicas diminuindo a efetividade da resposta imunológica (Jain 1993, Muller et al. 1994, Paes et al. 2000.O principal hormônio associado a condição estressante é o cortisol , que dependendo de seus teores séricos pode causar o leucograma caracterizado por neutrofilia, linfopenia, acompanhada ou não por alterações nos valores de monócitos, eosinófilos e contagem total de leucócitos, com redução da fagocitose e das funções oxidativas dos neutrófilos (Jain 1993, Raidal et al. 1997. Effect of stress on viral–bacterial synergy in bovine respiratory disease: novel mechanisms to regulate inflammation

The severity of bovine respiratory infections has been linked to a variety of factors, including environmental and nutritional changes, transportation, and social reorganization of weaned calves. Fatal respiratory infections, however, usually occur when a primary viral infection compromises host defences and enhances the severity of a secondary bacterial infection. This viral–bacterial synergy can occur by a number of different mechanisms and disease challenge models have been developed to analyse host responses during these respiratory infections. A primary bovine herpesvirus-1 (BHV-1) respiratory infection followed by a secondary challenge with Mannheimia haemolytica results in fatal bovine respiratory disease (BRD) and host responses to these two pathogens have been studied extensively. We used this disease model to demonstrate that stress significantly altered the viral–bacterial synergy resulting in fatal BRD. Functional genomic analysis revealed that BHV-1 infection enhanced toll-like receptors (TLR) expression and increased pro-inflammatory responses which contribute to the severity of a Mannheimia haemolytica infection. TLRs play a critical role in detecting bacterial infections and inducing pro-inflammatory responses. It is difficult to understand, however, how stress-induced corticosteroids could enhance this form of viral–bacterial synergy. Nuclear translocation of the glucocorticoid receptor activates cell signalling pathways which inhibit both TLR signalling and pro-inflammatory responses. The apparent conundrum between stress-induced corticosteroids and enhanced BRD susceptibility is discussed in terms of present data and previous investigations of stress and respiratory disease.
