Influence of MXene Particles with a Stacked-Lamellar Structure on Osteogenic Differentiation of Human Mesenchymal Stem Cells

Abstract: MXenes with a two-dimensional (2D) structure have attracted attention as potential biomedical materials. In this study, Ti3C2 MXene particles with 2D-lamellar structures were intercalated and their potential as a biomaterial was evaluated using human mesenchymal stem cells. Intercalated MXene was characterized in terms of microstructure, phase composition, and size. Cell proliferation experiments with MXene particles confirmed that concentrations >50 μg/mL were cytotoxic, while concentrations <20 μg/mL p… Show more

“…Treating human mesenchymal stem cells (hMSCs) with a high dose of Ti 3 C 2 T x nanosheets (>50 μg mL −1 ) led to obvious cytotoxicity, as shown in [ 37 ]. At a dose of 100 μg mL −1 , hMSCs displayed a significant decrease of cell proliferation early in this study.…”

“…Moreover, the mass ratio of MXene NPs in the nanofibrous matrices was carefully selected and used for the electrospinning process, taking into account the volume of the polymer solution and the results of the nanofibrous sheet (i.e., disc shape with 9 mm diameter). Based on the previous reports [ 13 , 35 ], we assumed that the mass ratio of the MXene NPs in the defined area was within the range of the optimized mass ratio evaluated by a series of the experiments, such as cytotoxicity and ALP activity tests. With a similar but slightly different from the previous approach, we also postulated that the MXene NPs in the nanofiber matrices may act differently on the cell behaviors because they are embedded in the surface of the matrices rather than placed on the cell culture plates.…”

“…Researchers assessed the osteogenic activity of MXene on human Mesenchymal Stem Cells (hMSCs) with cell culture medium containing different concentrations of MXene. 92 When the concentration was <20 μg/ml, hMSCs showed distinct proliferation and osteogenic differentiation activity. Moreover, the rising expression of early osteogenic genes RUNX2 and OPN and late osteogenic gene BSP was also observed under the stimulation of MXene.…”

“…To date, many scholars have made numerous efforts in MXene nanomaterials mediating the proliferation and early differentiation of cells. Researchers assessed the osteogenic activity of MXene on human Mesenchymal Stem Cells (hMSCs) with cell culture medium containing different concentrations of MXene 92 . When the concentration was <20 μg/ml, hMSCs showed distinct proliferation and osteogenic differentiation activity.…”
