Influence of Cold Plasma Atmospheric Jet on Surface Integrin Expression of Living Cells

Abstract: The effects induced in cells due to treatment with cold atmospheric plasma jet are studied. Cell migration rate is measured by means of time‐lapse microscopy. In order to characterize cell surface integrin expression, the fluorescent response of cells after surface integrins are stained with specific antibodies is measured by flow cytometry. We show that treatment of cells with plasma jet affects the cells on sub‐cellular level, namely decreases expression of cell surface integrins (β1 and αv integrins were te… Show more

“…Our experiments demonstrated that through adjustment of resistance and output voltage of CAP jet, with the change of plasma emission intensity and power, a mild dose with suitable discharge power can be acquired to efficiently kill metastatic BrCa cells while keeping healthy MSC alive. Due to the observed significantly inhibited migration of BrCa cells under CAP treatment, one of the possible effects of moderate intensity and power plasma treatment may contribute to alter BrCa cell surface receptor functions [24] (such as integrin mediating cell adhesion and migration) [25]. Furthermore, the present research demonstrated that CAP treatment plays an important role in tumor therapy via immediate cell lyses [26].…”

“…The kINPen 09, a plasma jet working with argon, caused no change in α 2 -integrin expression on HaCaT cells treated in suspension, while that of β 1 -integrin increased [21]. The expression of α V - and β 1 -integrin on adherent fibroblasts treated with a plasma jet using helium as working gas was downregulated [24]. In human melanoma cells treated with helium plasma the expression of α 2 - und α 4 -integrin was decreased [46].…”

“…ROS in this context are generated by cold plasma. Secondly, it could involve integrin-based activation by cold plasma, as also reported elsewhere 26 .…”

“…Although the mechanism of the interaction between cold plasma and cells or living tissue is still unclear 21 , several studies have reported the effects of cold plasma on key wound-related cells or sub-cells, included promoting theproliferationoffibroblasts 22 and endothelial cells 23 , as well as the growth of epithelial cells 24 , inhibiting the migration of fibroblasts 25 and their surface expression 26 , and activating integrin of fibroblasts and epithelial cells 27 . Interestingly, some of these effects are likely to be similar to the activities of natural ROS and/or NO during wound healing, particularly cold plasma's effects on the proliferation of both fibroblasts and endothelial cells 19 .…”
