In vitro stability and expression of green fluorescent protein under high pressure conditions

Abstract: Aims: The objective of this work was to evaluate the use of wild‐type GFP and mutant forms thereof as reporter for gene expression under high pressure conditions. Methods and Results: The intensity of fluorescence after high pressure treatment was checked by subjecting cells, crude protein extracts containing GFPs and purified GFPs to pressures ranging from 100 MPa to 900 MPa. All tested GFP’s retained fluorescence up to 600 MPa without loss of intensity. Expression of GFP under sublethal conditions was inves… Show more

“…The protein is in the shape of a cylinder named β-can, consisting of 11 strands of β-sheet with an α-helix inside, which protect the fluorophore [49,50]. GFP is quite thermostable [51], very resistant to chemical and pressure-induced denaturation [52][53][54].…”

“…4) Pressure variations are immediately reported in the fluorescence spectrum. 5) The protein stability domain, which ranges up to at least 600 MPa [53,54], exceeds the pressure domain that is used in experiences performed with most live microorganisms or their cellular enzymatic machinery.…”

“…The wide-range use of GFP is a consequence of its unique properties: reduced sensitivity to higher temperatures, detergents, alkaline pH, photobleaching, organic salts, chaotropic salts and many proteases [10]. EGFP (enhanced green fluorescent protein, a mutant version of GFP) has emerged as a powerful fluorescent label for quantitative fluorescence microscopy applications [11].…”

“…Environmental stress encountered by E. coli in the human intestine or in food processing also induces Stx-prophages (18)(19)(20). Human neutrophils in the intestine release antibacterial molecules, including H 2 O 2 , which stimulate Stx and phage production (19).…”
