In Vitro Simultaneous Transfer of Lipids to HDL in Coronary Artery Disease and in Statin Treatment

Abstract: The exchange of lipids with cells and other lipoproteins is a crucial process in HDL metabolism and for HDL antiatherogenic function. Here, we tested a practical method to quantify the simultaneous transfer to HDL of phospholipids, free-cholesterol, esterified cholesterol and triacylglycerols and to verify the lipid transfer in patients with coronary artery disease (CAD) or undergoing statin treatment. Twenty-eight control subjects without CAD, 27 with CAD and 25 CAD patients under simvastatin treatment were s… Show more

“…PON1 activity is substantially lower in patients with CAD, ACS, CKD, and RA, whereas lipoprotein-associated phospholipase A 2 (LpPLA 2 ) activity is increased in CKD and psoriasis (89,137,160,165,(171)(172)(173)(174)(175). Dysregulating the activity of PON1 and LpPLA 2 in this fashion not only compromises the antioxidative activity of HDL, but also promotes the formation of atherosclerotic plaques.…”

“…The transfer rates of cholesteryl ester, phospholipid, free cholesterol and triglyceride from cholesterol-rich emulsions to HDL was determined in the Lipid Metabolism Laboratory of the Heart Institute (InCor) as previously described (Daminelli et al, 2008;Giribela et al, 2009;Lo Prete et al, 2009;Puk et al, 2009). Lipid nanoemulsions were prepared from a lipid mixture of 40 mg cholesteryl oleate, 20 mg egg phosphatidylcholine, 1 mg triolein and 0.5 mg cholesterol purchased from the Sigma Chemical Company (St Louis, MO, USA).…”

“…ferent (p < 0.05) when compared to normolipidemic group. The fact that the participating individuals did not use lipid-lowering drugs at the time of sampling was important to exclude treatment interference on lipid metabolism, a really important aspect not found in several important published studies [18] [19].…”
