Modeling and comparison of the structures of the two closely related cutinases Thc_Cut1 and Thc_Cut2 from Thermobifida cellulosilytica DSM44535 revealed that dissimilarities in their electrostatic and hydrophobic surface properties in the vicinity to the active site could be responsible for pronounced differences in hydrolysis efficiencies of polyester (i.e., PET, polyethyleneterephthalate). To investigate this hypothesis in more detail, selected amino acids of surface regions outside the active site of Thc_Cut2, which hydrolyzes PET much less efficiently than Thc_Cut1 were exchanged by site-directed mutagenesis. The mutants were expressed in E. coli BL21-Gold(DE3), purified and characterized regarding their specific activities and kinetic parameters on soluble substrates and their ability to hydrolyze PET and the PET model substrate bis(benzoyloxyethyl) terephthalate (3PET). Compared to Thc_Cut2, mutants carrying Arg29Asn and/or Ala30Val exchanges showed considerable higher specific activity and higher kcat /KM values on soluble substrates. Exchange of the positively charged arginine (Arg19 and Arg29) located on the enzyme surface to the non-charged amino acids serine and asparagine strongly increased the hydrolysis activity for 3PET and PET. In contrast, exchange of the uncharged glutamine (Glu65) by the negatively charged glutamic acid lead to a complete loss of hydrolysis activity on PET films. These findings clearly demonstrate that surface properties (i.e., amino acids located outside the active site on the protein surface) play an important role in PET hydrolysis.

Silver‐doped hydroxyapatite (AgHAp) was prepared by co‐precipitation method at room temperature. The obtained AgHAp was added in different amounts of collagen gel (AgHApC1 and AgHApC2). Afterward, the gel was lyophilized and the final AgHApC1 and AgHApC2 composite was achieved. The purity, crystallinity, and the phase composition of the AgHAp, AgHApC1, and AgHApC2 samples were evaluated by X‐ray diffraction (XRD). The planes corresponding to the 2θ values for hydroxyapatite were found in the three samples analyzed in agreement with the crystalline hydroxyapatite. In the Fourier transform infrared (FT‐IR) spectra of AgHApC1 and AgHApC2 samples the peak characteristic to the presence of ν2 phosphate mode at 472/cm was found. The peaks resulting from the ν4 vibration of the P–O mode, ν1 symmetric P–O stretching vibration and ν3 P–O stretching vibration of PO43− were also evidenced in all the samples. The formation of agglomerated particles with uniform particle size was evidenced by scanning electron microscope (SEM). The uniform distribution of the constituent elements was evidenced by mapping analysis. Furthermore, the strong antibacterial activity of AgHAp, AgHApC1, and AgHApC2 samples against Gram‐negative (Escherichia coli) and Gram‐positive (Staphylococcus aureus) bacterial strains was shown. Surgical transaxillary placement of the Impella 5.0 ventricular assist device

Objective The aim of this study is to evaluate the open transaxillary placement of the Impella 5.0 with a modified surgical technique. Methods From January to July 2018, nine patients (eight males; mean age 60 years) underwent surgical transaxillary Impella 5.0 (Abiomed Inc., Danvers, MA) implantation. Patient and periprocedural data were recorded in a prospectively maintained institutional database. The primary endpoint was technical success. The secondary endpoints were: neurological complications (peripheral or central), bleeding, and wound infection, duration of Impella support, and device failure requiring device replacement. Results Assisted technical success was 100%. The right axillary artery was used in 8/9 cases. Three patients (all on extracorporeal membrane oxygenation) suffered from access site bleeding which required surgical reintervention. One patient suffered from peripheral neurological dysfunction which recovered in 1 month and one patient had a local hematoma which was managed conservatively. The median length of treatment was 16 days (range 8‐35). Five patients had myocardial recovery and the Impella could be explanted; the remaining were transitioned to a durable left ventricular assist device with an uneventful postoperative course. Hospital mortality was 33%. Conclusions Open Impella 5.0 device implantation through the axillary artery is safe and effective; technical success was 100% and low rates of major complications are reported.
