Immunohistochemistry: sole tool in diagnosing a rare case of primary vaginal amelanotic melanoma

Abstract: We report a rare case of vaginal amelanotic melanoma. Malignant melanomas are cutaneous and extracutaneous tumors that arise from embryological remnants of neural crest cells/melanocytes. Amelanotic melanomas at such rare locations can be misdiagnosed both clinically and radiologically. Therefore, histopathological examination and immunohistochemistry are mandatory for the diagnosis of these tumors. We diagnosed this case using histopathology and confirmed the diagnosis based on the presence of immunohistochem… Show more

“…On immunohistochemistry tumor cells were strongly positive for HMB45, vimentin and S-100. 12 In another study by Khairwa et al, microscopic findings showed an infiltrating tumour in the surrounding tissue, arranged in sheets, nests and lobules with atypical cells showing moderate pleomorphism with abundant eosinophilic cytoplasm and frequent mitosis. On immunohistochemistry, the tumour cells showed positivity for HMB45 and S-100.…”

“…Histolopathological findings of our study correlated well with the other studies. 3,[11][12][13] Microscopic examination showed sheets, cords and singly scattered markedly pleomorphic, polygonal with eccentrically placed nuclei with conspicuous nucleoli along with eosinophiliccytoplasm. Focal intranuclear inclusions and frequent mitosis was also seen.…”

“…The etiology of AMM remains unclear, although it was previously hypothesized that decreased tyrosinase activity and/or melanin transport disorders may constitute possible reasons ( 11 ). AMM has been reported in the skin and the mucosa of the digestive and/or genitourinary tracts ( 1 , 10 ). AMM is a clinically diverse entity without specific characteristics; early lesions are atypical and usually appear as pinkish papules, either as a single lesion or in clusters ( 10 , 12 ).…”

“…Malignant melanoma (MM) is a cutaneous and/or extracutaneous tumor that arises from the embryological remnants of neural crest cells/melanocytes, melanoma can happen in eyes and internal organs but rare ( 1 ). The number of recorded new cases in 2018 was 2,87,723, whereas the number of deaths in 2018 in both sexes and across all ages was 60,712 (Globocan 2018, International Agency for Research on Cancer, World Health Organization; https://gco.iarc.fr/today/data/factsheets/cancers/16-Melanoma-of-skin-fact-sheet.pdf ).…”
