Immunoblot evaluation of the 100 and 130 kDa antigens in camel hydatid cyst fluid for the serodiagnosis of human cystic echinococcosis in Libya

Abstract: Two high molecular weight antigens with molecular masses approximately 100 kDa and 130 kDa were identified by immunoblotting camel hydatid cyst fluid, with 94% sensitivity in sera from surgically confirmed Libyan cystic echinococcosis cases. 40% of sera from surgically confirmed alveolar echinococcosis cases cross-reacted with the 100 and 130 kDa antigens, as did 5.3% of sera from human Taenia solium cysticercosis patients. No cross-reaction occurred with sera from human schistosomiasis mansoni or onchocercias… Show more

“…The sensitivity (92%) for the 8-kD subunit obtained in our study appears to be similar to that (94%) for the 8-kD subunit reported by Shambesh and others, 11 as well as that (90%) for the 12-kD subunit described by Leggatt and others, 10 but higher than that (80%) for the 12-kD subunit reported by Ioppolo and others. 31 The cross-reactions for the 8-kD subunit with AE sera (80%) demonstrated in our study is higher than those reported in other two studies (both 40%).…”

“…31 The cross-reactions for the 8-kD subunit with AE sera (80%) demonstrated in our study is higher than those reported in other two studies (both 40%). 10,11 Based on these data, it seems reasonable to conclude that the smallest subunit of antigen B (8/12-kD) is not species-specific for E. granulosus but is shared with E. multilocularis. This will limit its usefulness for serologic differentiation of CE from AE.…”

“…Some of the products identified in HCF by the pool of human sera in the present study may correspond to those previously described by other authors using the same antigenic extract. This may be the case of the protein of 34 kDa (recently identified by Poretti et al 1999), and the protein of 110 kDa, probably related with the component of 100 kDa described by Shambesh et al (1995), the protein of 110-120 kDa (Shapiro et al 1992), and the protein of 116 kDa (Kanwar et al 1992).…”

“…According to the ROC curve analysis, the band of 120-124 kDa is observed as the most important band for differentiation. There aren't enough studies about high molecular masses in CE patients (Sbihi et al, 1997;Kanwar et al, 1992;Shambesh et al, 1995). In a previous study, we frequently found a band of 124 kDa in patients with pulmonary CE (Akisu et al, 2005).…”
