Immuno Affinity SELEX for Simple, Rapid, and Cost-Effective Aptamer Enrichment and Identification against Aflatoxin B1

Abstract: Aflatoxins are naturally occurring mycotoxins that contaminate food and agro commodities, leading to acute and chronic health conditions in human and animals. In the present work, an attempt was made to generate high-affinity single stranded DNA aptamers that specifically bind to Aflatoxin B1 (AFB1) by a modified Systemic Evolution of Ligands by Exponential Enrichment (SELEX) procedure with the aid of Immunoaffinity columns. Ten rounds of SELEX and alternating three counter SELEX rounds with a cocktail of rela… Show more

“…Compared to traditional SELEX, counter SELEX adds an additional step using structurally-similar targets to incubate with aptamers to effectively discriminate non-specific oligonucleotides. This procedure has been dramatically applied to other modified SELEX methods to obtain more specific aptamers [ 10 , 11 , 12 ]. It is to be noted that the prominent difference between counter SELES and negative SELEX is their use of different incubation objects.…”

“…In early 2015, a heightened risk of aflatoxin poisoning became a concern of the Provincial Government of East Java, Indonesia, due to the increase in small industries which commercially process and market animal feed traditionally without considering sanitation and/or product quality [ 1 ].…”

“…Since its launch in 1990, conventional SELEX has been progressively modified to improve aptamer specificity and affinity, to simplify the process and to increase time and cost efficiency [ 44 46 ]. The modifications in the SELEX method depend on the purpose and target molecules.…”
