IL1R2 Blockade Suppresses Breast Tumorigenesis and Progression by Impairing USP15‐Dependent BMI1 Stability

Abstract: Breast tumor initiating cells (BTICs) with ALDH+CD24−CD44+ phenotype are the most tumorigenic and invasive cell population in breast cancer. However, the molecular mechanisms are still unclear. Here, it is found that a negative immune regulator interleukin‐1 receptor type 2 (IL1R2) is upregulated in breast cancer (BC) tissues and especially in BTICs. BC patients with high IL1R2 expression have a poorer overall survival and relapse‐free survival. High IL1R2 promotes BTIC self‐renewal and BC cell proliferation a… Show more

“…However, there is less information on the association of IL1R2 with IL-1β-dependent intracellular signaling during the pathogenesis of diseases. The increased expression of IL1R2 has been associated with poor prognosis and metastasis of several malignancies [ 23 , 24 ]. The ratio of IL-1β/IL1R2 positively correlated with the production of interferon γ, and negatively correlated with TNF-α production in women with invasive ductal mammary adenocarcinomas [ 23 ].…”

“…B cell-specific Moloney murine leukemia virus integration site 1 (BMI1) is suggested to undergo upregulation in different cancers. BMI1 overexpression ensures tumor sphere formation of cancer cells, and its stability is a positive factor for tumorigenesis [ 104 , 105 ]. MiR-15 as an onco-suppressor factor has been reported to inhibit EMT in BC cells.…”

“…IL-1R2 is mainly expressed in neutrophils, B cells, monocytes, and macrophages. Plasma levels of soluble IL-1R2 are between [5][6][7][8][9][10] ng/ml in healthy donors and increase in patients with infectious conditions [7] . IL-1R2 plays a role in a variety of diseases, including chronic skin inflammation, arthritis, endometriosis, and heart transplantation or autoimmune myocarditis [7] .…”
