IgGs-Abzymes from the Sera of Patients with Multiple Sclerosis Recognize and Hydrolyze miRNAs

Abstract: Autoantibodies-abzymes hydrolyzing DNA, myelin basic protein, and oligosaccharides have been revealed in the sera of patients with multiple sclerosis (MS). In MS, specific microRNAs are found in blood and cerebrospinal fluid, which are characterized by increased expression. Autoantibodies, specifically hydrolyzing four different miRNAs, were first detected in the blood of schizophrenia patients. Here, we present the first evidence that 23 IgG antibodies of MS patients effectively recognize and hydrolyze four n… Show more

“…It showed earlier that all IgGs and sIgAs effectively hydrolyze four microRNAs: miR-9-5p, miR-219-2-3p, miR-137, and miR-219a-5p, which are found in high concentrations in the blood of patients with autoimmune diseases [ 73 , 74 ]. Elevated concentrations of these four microRNAs are more characteristic of patients with SLE and MS, and IgGs effectively hydrolyzed them from the blood of these patients [ 69 , 70 , 71 , 72 ].…”

“…The spatial models of four microRNAs were generated by Predict a Secondary Structure server, which uses a combination of four algorithms for predicting the secondary structure of RNA, similarly to [ 69 , 70 , 71 ]: calculating a partition function, denoting the structure with minimal energy by (accessed on 14 February 2018).…”

“…This reflects the peculiarity of the immunological reactivity of the organism during this disease. Autoantibodies-abzymes-that hydrolyze microRNAs were found in the sera of patients with multiple sclerosis and systemic lupus erythematosus [111,112]. Evidence has been presented that the secretory IgA molecules of breast milk (but not the blood sera of healthy men and non-pregnant women) can effectively hydrolyze ribooligonucleotides, i.e., they are abzymes that effectively hydrolyze microRNAs in their loop and duplex regions.…”

“…One of the specific features of AI disease progression was shown to be the appearance of antibodies (abzymes) in the patients’ blood that hydrolyze DNA, RNA, proteins, nucleotides, and oligosaccharides (for review, see [ 6 , 7 , 8 , 9 , 10 , 11 , 12 ]). The blood of MS patients was found to contain abzymes that efficiently hydrolyze DNA [ 13 , 14 ], RNA, specific microRNA [ 15 ], the myelin basic protein [ 16 , 17 , 18 ], five histones [ 19 ], and oligosaccharides [ 20 , 21 ]. Interestingly, IgG antibodies from the cerebrospinal fluids of MS patients were 30–60 times more active in hydrolysis of MBP, DNA, and oligosaccharides than from the serum of the same patients [ 22 , 23 , 24 ].…”
