Identification and comparison of three carp fishes based on mitochondrial 16S rRNA gene

Abstract: Identification of Labeo rohita, L. bata and L. gonius is sometimes problematic when usual morphological features are lost and it is difficult to differentiate them with traditional morphological features at their diverse developmental stages. PCR-sequencing provides an authentic alternative means of identification of individuals at species level. Three local carp fishes were collected and 16S rRNA genes were sequenced by sanger sequencing method after PCR amplification using universal primers. Obtained sequenc… Show more

“…In molecular identification, mitochondrial COI, 16SrRNA, 12SrRNA and cyt-b are used frequently. Jahan et al (2017) have studied 16SrRNA gene sequence of three Labeo species viz., L. rohita, L. bata and L. gonius and found 23 polymorphic sites (5.8%) in 396 nucleotide long sequence. The polymorphic site variations (23.5%) are more frequent than that reported by Jahan et al (2017) because of larger size sequences, higher number of species (eight) and use of different region of mitochondrial genome (part of cyt-b) in the present study.…”

“…This is due to the use of 16S mitochondrial DNA as a molecular marker has several advantages such as maternal mode of inheritance, limited recombination, and high conserve which can be used to study species identification and interspecies relationship [8]. The use of 16S mitochondrial marker had been successfully applied to identify species identification [9][10][11].…”

“…Therefore, in this study we use partial 16S mitochondrial gene as a molecular marker to detect polymorphism in the spotted barb fish. The 16S mitochondrial gene has been widely used to identify and detect genetic variation of freshwater fish such as carp fishes (Jahan et al, 2017) and kissing gourami (Arisuryanti et al, 2019 a ), Serranidae fish (Saad, 2019), and red sea parrotfish (Saad et al, 2019). The finding data can be also utilised to arrange 16S mt-DNA library sequences of the spotted barb fish from Indonesia.…”
