“…The spleen leucocytes were segregated from Nile Tilapia by Histopaque ® 1077 (Sigma, USA) as previous reports (Mu et al, ; Wu et al, ). Simply, spleen was grinded in a flask, stacked on top of 1077 and centrifuged.…”

“…The leucocytes were counted and re‐suspended with/L15 medium, containing 1% penicillin/streptomycin (Sigma, USA) and 10% foetal bovine serum (FBS) (Gibco, USA). The cells were cultured in 96‐well plate (Corning, USA; 100 μl/well) with the final concentration of 1 × 10 6 cells/ml per well and then stimulated with poly (I:C) (50 μg/ml), S. agalactiae (1 × 10 7 CFU/ml) (Mu et al, ; Wang, Seymour, Sundberg, & Hogenesch, ) and A. hydrophila (1 × 10 7 CFU/ml) at 25℃. Bacteria of S. agalactiae and A. hydrophila (Tang et al, ; Yin et al, ) were cultured and inactivated and control group with the equal volume of 1 × PBS as same as our previous study.…”

“…Protein conformation is influenced by the ions present, temperature, and pH that specifically influences the agglutination capacity of the proteins. Therefore, heating can inhibit agglutination [ 27 ] and agglutination can be Ca ++ dependent [ 27 , 28 ]. Lectins that only have one carbohydrate recognising domain (CRD) must form dimers or higher-order structures to be able to agglutinate.…”
